A concise synthesis of novel spiroisoxazol-5-one-tetrahydroquinoline derivatives e.g., I (X = O) was developed by zinc chloride-catalyzed stereoselective Knoevenagel condensation/1,5-hydride shift/cyclization reaction of o-(dialkylamino)benzaldehydes with isoxazol-5-ones. Using this method, spiroisoxazolone-tetrahydroquinoline products were obtained in good to high yields (up to 97 % yield) with good to excellent diastereoselectivities (up to >95:5 dr). In addition, a novel 5-hydroxy-spiroisoxazoline derivative II was prepared by reduction of compound I (X = O) with lithium aluiminum hydride. Furthermore, spiroisoxazolone-tetrahydroquinoline derivative I (X = O) was transformed into spiropyrazolone I (X = NH) by reaction with hydrazine hydrate.
