General procedure: Ethyl 5-(hydroxymethyl)isoxazole-3-carboxylate (prepared according to WO2010/142801, p. 147, 0.093 g) was dissolved in dichloromethane (5 ml) and 1,1′-carbonyldimidazole (0.089 g) was added. The reaction mixture was stirred for 2 h at room temperature before (2E,4E)-7-butoxyhepta-2,4-dien-1-amine hydrochloride (0.120 g) and triethylamine (0.076 ml) were added. The reaction mixture was warmed to 40C and stirred overnight after which it was cooled to room temperature and washed with water. The organic layer was collected and evaporated in vacuo. The crude product was purified via flash column chromatography to yield the product as a white solid

Step A – (5-Hydroxymethyl-isoxazol-3-yl)-diphenyl-methanol; A solution of triethylamine (69 ml_) in ether (31 mL) was added slowly over 4 h with the aid of a syringe pump to a briskly stirred solution of propargyl alcohol (37.5 mL) and ethyl 2-chloro-2-(hydroxyimino)acetate (75 g) in ether (500 mL) at room temperature. The reaction mixture was then allowed to stir overnight, filtered and the filtrate washed with water (twice). The aqueous phases were combined, saturated with sodium chloride and re-extracted with ethyl acetate (twice). The combined organic phases were dried (MgSO4), filtered and evaporated in vacuo to give a thick oil (82 g) comprised mainly of delta-hydroxymethyl-isoxazole-S-carboxylic acid ethyl ester. This was dissolved in THF (700 mL), cooled to -10DC and treated with a solution of phenylmagnesium chloride (750 mL, 2.0 M in THF) keeping the temperature below -2C. The reaction mixture was warmed to room temperature and stirred for 1 h. The reaction mixture was poured carefully into ice-cold concentrated hydrochloric acid (200 mL) and ice (500 mL) and the layers separated. The aqueous layer was extracted with ether. The combined organic layers were washed with brine, dried, filtered and evaporated in vacuo. 0.172 g (1.00 mmole) of ethyl 5-hydroxymethyl-isoxazole-3-carboxylate and 0.33 ml (1.92 mmoles) of N,N-diisopropylethylamine are dissolved in 9 ml of 1,2-dichloroethane then cooled to 0 C. 0.184 g (0.91 mmole) of p-nitrophenyl chloroformate in solution in 2 ml of 1,2-dichloromethane are added. The mixture is stirred for 20 mins at ambient temperature, then 0.230 g (0.91 mmole) of 2-(4-chlorophenoxy)-7-aza-spiro[3.5]-nonane, obtained in stage 2.2, is added. The mixture is heated at 60 C. for 15 hrs. After return to ambient temperature, a 1N aqueous solution of caustic soda is added, and the product is extracted with dichloromethane. The combined organic phases are then successively washed three times with a 1N aqueous solution of caustic soda, twice with a saturated aqueous solution of ammonium chloride and once with a saturated aqueous solution of sodium chloride, dried over sodium sulphate, filtered and evaporated to dryness. 0.447 g of the expected product are obtained in the form of a colourless oil which is used as such in the following stage., 123770-62-7
