Design, synthesis and antitrypanosomatid activities of 3,5-diaryl-isoxazole analogues based on neolignans veraguensin, grandisin and machilin G

Using bioisosterism as a medicinal chemistry tool, 16 3,5-diaryl-isoxazole analogues of the tetrahydrofuran neolignans veraguensin, grandisin and machilin G were synthesized via 1,3-dipolar cycloaddition reactions, with yields from 43% to 90%. Antitrypanosomatid activities were evaluated against Trypanosoma cruzi, Leishmania (L.) amazonensis and Leishmania (V.) braziliensis. All compounds were selective for the Leishmania genus and inactive against T. cruzi. Isoxazole analogues showed a standard activity on both promastigotes of L. amazonensis and L. braziliensis. The most active compounds were 15, 16 and 19 with IC50 values of 2.0, 3.3 and 9.5 mu M against L. amazonensis and IC50 values of 1.2, 2.1 and 6.4 mu M on L. braziliensis, respectively. All compounds were noncytotoxic, showing lower cytotoxicity (>250 mu M) than pentamidine (78.9 mu M). Regarding the structure-activity relationship (SAR), the methylenedioxy group was essential to antileishmanial activity against promastigotes. Replacement of the tetrahydrofuran nucleus by an isoxazole core improved the antileishmanial activity.

Polymorphic and solvate structures of ethyl ester and carboxylic acid derivatives of WIN 61893 analogue and their stability in solution

3-Ethyl ester- (1) and 3-carboxylic acid-isoxazole (2) derivatives of an antiviral drug analogue WIN 61893 were synthesized and characterized by X-ray crystallography and NMR spectroscopy. Crystallization experiments afforded two polymorphic structures for the ethyl ester derivative and two solvate structures for the carboxylic acid derivative based on their ability to form intermolecular hydrogen bonding interactions with the solvent molecules. The conformations of the derivatives depended greatly on the orientation of the planar isoxazole and phenyl-oxadiazole ring systems with respect to one another and were found to take up perpendicular, linear or tilted conformations. The carboxylic acid derivative was furthermore observed to undergo isoxazole ring cleavage by decarboxylation in DMSO solution and transforming into a beta-keto nitrile ring-opening product over several days, whereas, the isoxazole ring of the ethyl ester derivative was not affected.

The synthesis of the first examples of the imidazo[4,5-c]isoxazole ring system has recently been reported,(2) but little is known about the chemistry of this heterocycle. In this paper we describe our investigation into the behaviour of an imidazo[4,5-c]isoxazole-6-carboxylate ester with acetylenic esters and ketones. A complex reaction sequence is found to occur, involving addition of two molecules of alkyne followed by ring opening and fragmentation, leading to the formation of 2-pyrrol-2-yl imidazoles in moderate yield. Synthesis, biological evaluation and docking study of some novel isoxazole clubbed 1,3,4-oxadiazoles derivatives

A novel series of isoxazole clubbed 1,3,4-oxadiazole derivatives have been synthesized by reaction of 5-(3-fluoro-4-methoxyphenyl) isoxazole-3-carbohydrazide with different substituted benzoic/pyridinyl/indolyl acids in phosphorous oxychloride, characterized by IR, H-1 NMR, C-13 NMR, MS analytical data and evaluated for their antimicrobial as well as antitubercular activity. Antibacterial activity of compounds 5e, 5g, 5h, 5j and 5l were found to be good against E. coli, P. aeruginosa, S. aureus and S. pyogenes as compared to standard Ampicillin. Compound 5b and 5i were found to be active antitubercular agents against M. tuberculosis H37Rv. Antibacterial and antitubercular activity was supported by molecular docking to gain insights of the mode of inhibition of MurD ligase enzyme.
