EXAMPLE 78 – PREPARATION OF Lambda/-(2-(5-chloro-1H-indol-3-yl)ethyl)-3-phenylisoxazole- 5-carboxamide.; A solution of lithium hydroxide 2 M in water (3 mL) was added to a mixture of the intermediate 54 (ethyl 3-phenylisoxazole-5-carboxylate) (0.200 g; 1.18 mmol) in ethanol (2 mL). The mixture was stirred vigorously at room temperature overnight and concentrated under reduced pressure. The pH was adjusted to 1 with HCI 6N, and the aqueous layer was extracted with ethyl acetate. The organic layer was dried over magnesium sulfate, filtered and evaporated to dryness under reduced pressure to afford the desired crude carboxylic acid which was directly engaged in the next step.A solution of the crude acid, 2-(5-chloro-1 /-/-indol-3-yl)ethanamine hydrochloride (0.298 g; 1.29 mmol), HATU (0.536 g; 1.41 mmol) and Lambda/,Lambda/-Diisopropylethylamine (0.506 mL; 2.94 mmol) in DMF (5 mL), was stirred at room temperature for 72 hours. The reaction mixture was concentrated under reduced pressure, diluted in ethyl acetate, and washed with water. The organic layer was dried over magnesium sulfate, filtered and concentrated under reduced pressure. The crude material was purified by flash chromatography on silica gel (eluent 20 to 80% ethyl acetate in heptane) to afford 0.0146 g ( 3%) of Lambda/-(2-(5- chloro-1 /-/-indol-3-yl)ethyl)-3-phenylisoxazole-5-carboxamide. ESI/APCI(+): 379 (M+H). ESI/APCK-): 377 (M-H).
