A series of 18 chromone derivatives I [R = HCC(O)OCH2CH3, 2,4,6-trioxo-1,3-diazinan-5-yl, 5-oxo-3-phenyl-4,5-dihydro-1,2-oxazol-4-yl, 1,3-dioxo-2,3-dihydro-1H-inden-2-yl, piperidin-1-amine, etc.] were designed and synthesized from paeonol. All synthetic compounds I were evaluated for their anti-proliferative activity towards eight human cancer cell lines including HepG2, HL-60, KB, LLC, LNCaP, LU-1, MCF7, and SW480. The compounds I (R = 2,4,6-trioxo-1,3-diazinan-5-yl, 1,3-dimethyl-2,4,6-trioxo-1,3-diazinan-5-yl) presented the best cytotoxic effects towards tested cancer cell lines except HepG2. Their IC50 values were ranging from 7.9±0.4 to 18.9±1.3 μg/mL. Meanwhile, compound I (R = 2-imino-3-methyl-5-oxoimidazolidin-4-yl) showed selective cytotoxicity against MCF7 and KB with IC50 of 13.7±0.6 and 15.5±0.9 μg/mL, resp.
