Objective To observe the effects of early use of parecoxib sodium on serum tumor necrosis factor-α(TNF-α) in mice with burn. Methods Two hundreds ICR male mice were randomly divided into 4 groups with 50 mice each. At 4 h after the burn models were established, the mice were i.p. injected with normal saline (group IU), parecoxib sodium 0.75 mg/kg (group TA) or parecoxib sodium 7.5 mg/kg (group TB), which was repeated at 24 and 48 h after the first injection. The mice without burn were taken as the controls (group U). Serum levels of TNF-α were detected by CBA liquid chip method at 6(T1), 12(T2), 24(T3), 48(T4) and 72 h (T5) after the first injection. Results Compared with group U, serum concentration of TNF-α in group IU at each time point was increased significantly (P<0.05). Serum level of TNF-α at T1 were higher in group TA than that in group U (P<0.05). Serum levels of TNF-α at T2-T5 were lower in group TB than those in group U (P<0.05). Serum levels of TNF-α at T2-T5 in group TA and at T1-T5 in group TB were lower than those in group IU (P<0.05). Conclusion The application of parecoxib sodium 0.75 mg/kg in the early stage of burn can moderately decrease serum concentration of TNF-α, which is beneficial to inhibiting inflammation in mice with burn. But the use of parecoxib sodium 7.5 mg/kg may decrease serum concentration of TNF-α excessively, which is not beneficial to keeping the rebalance of proinflammatory cytokines and anti-inflammatory cytokines.
