A simple isocratic procedure was developed for the anal. of new serotonergic anxiolytics and related compounds in bulk, pharmaceuticals, and in biol. samples. The system may be applied for the assay of other serotonergic anxiolytics of related structure such as buspirone. The HPLC assay utilized a reversed-phase C18 column, a mobile phase consisting of a mixture (55:45) of (A) buffer potassium dihydrogen phosphate (0.05M) containing sodium lauryl sulfate (0.005M) and (B) MeCN. A fluorescence detection was used with λex 237 nm; λem 374 nm. The accuracy, precision and sensitivity of the proposed method were established. Standard curves were linear with respect to concentration in the range 0.05-7.5 μg mL-1. The method also allowed the separation and identification of related compounds at concentrations <0.01%.
