Preparation of N-(1-benzyl-1H-pyrazol-4-yl)-5-cyclopropylisoxazole-3-carboxamide (119) To a solution of 1-benzyl-1H-pyrazol-4-amine (0.050 g, 0.289 mmol), 5-cyclopropylisoxazole-3-carboxylic acid (44.1 mg, 0.289 mmol) and 1-[bis(dimethylamino)methylene]-1H-1,2,3-triazolo[4,5-b]pyridinium 3-oxide hexafluorophosphate (109 mg, 0.289 mmol) in N,N’-dimethylformamide (1 mL) at 25 C. was added diisopropylethylamine (0.075 mL, 0.433 mmol). The reaction mixture was stirred at 25 C. for 16 h. The reaction mixture was quenched with water (1 mL). The aqueous layer was extracted with ethyl acetate (5 mL*3). The combined organic layers were washed with brine, dried over magnesium sulfate, filtered and concentrated in vacuo. The crude residue was purified by column chromatography (ISCO, 12 g silica, eluting with 40% ethyl acetate/hexanes for 20 min) to give N-(1-benzyl-1H-pyrazol-4-yl)-5-cyclopropylisoxazole-3-carboxamide (12.4 mg, 0.041 mmol, 14%) as an off-white solid. 1H NMR (300 MHz, Chloroform-d) delta 8.42 (s, 1H), 8.00 (s, 1H), 7.59 (s, 1H), 7.37-7.18 (m, 5H), 6.39 (s, 1H), 5.30 (s, 2H), 2.11 (tt, J=8.5, 5.0 Hz, 1H), 1.23-1.08 (m, 2H), 1.08-0.92 (m, 2H); LCMS (ESI) m/z: 309.2 [M+H]+.
