To a solution of 3-(5-(aminomethyl)-2-chlorophenyl)-1-(4-(trifluoromethyl)phenyl)-1H- 1,2,4-triazol-5(4H)-one (Intermediate-63, 0.060 g, 0.162 mmol) in dry THF ( 5 mL) was added DIPEA (3 mL) and stirred for 20 minutes. The reaction mixture was cooled to 0C and isoxazole-5-carbonyl chloride (0.032 g, 0.243 mmol) was added and stirred for 3 h at room temperature. The reaction mass was quenched in water, extracted with DCM: MeOH and concentrated to afford crude product which was purified by column chromatography eluting with MeOH: DCM to afford 0.030 g of pure product. 1H NMR (400 MHz, DMSO d6): delta 4.50-4.52 (d, J = 8Hz, 2 H), 7.09-7.10 (d, = 2 Hz, 1H), 7.43- 7.45 (d, J = 8.4 Hz, 1H), 7.50-7.53 (d, J = 10.4Hz, 1H), 7.59-7.64 (d, J= 18 Hz, 1H), 7.82-7.85 (d, / = 8.8 Hz, 2H), 8.17-8.19 (d, J = 8.4 Hz, 2H), 8.74-8.75 (d, J – 2Hz, 1H), 9.58-9.61 (t, 1H), 12.61-12.74 (br s, 1H); MS (m/z): 464.11 (M+H+).

62348-13-4, To a mixture of tert-butyl 4-(4-(2-chloropyrimidin-4-yi)-lH-pyrazol-l-yl)-4- (cyanomethyl)piperidine- 1 -carboxylate (0.259 g, 0.643 mmol) in 2 mL of dioxane was added 4 M HCl in dioxane (4.0 mL). The reaction was stirred at room temperature for 30 min, then evaporated to dryness. To the resulting crude HCl salt in methylene chloride (5.0 mL) was added triethylamine (0.269 mL, 1.93 mmol) followed by isoxazole-5-carbonyl chloride (0.0744 mL, 0.771 mmol). The mixture was stirred at room temperature for 30 min, washed with saturated sodium bicarbonate, dried, and evaporated to dryness. The residue was used directly in next step (233 mg, 91%). LCMS (M+H) 398.0.

62348-13-4, To PS-HOBT resin (0.1 mmol) was added anhydrous dichloromethane (“DCM”) (1 mL) followed by pyridine (0.5 mmol) and isoxazole-5-carbonyl chloride (Lancaster) (0.3 MMOL). The mixture was shaken at room temperature for 3 h and was then filtered. The resin was washed successively with tetrahydrofuran (“THF”) (3x) and DCM (3x) and dried III vacuo. To this acylated resin was added a solution of 2- piperidinoaniline (Lancaster) (0.05 mmol, 0.5 eq) in anhydrous THF (1 mL) and the mixture was shaken at room temperature for 16 H. THE MIXTUE WAS THEN FILTERED AND the resin washed with THF and DCM as described above. The combined filtrate and washings were concentrated under reduced pressure to yield the product. Yield: 100%. MS: 272 (M+1). LC/MS PURITY : 100%. IHNMR (CDCL3, 300MHZ) : 58. 2 (d, 1H), 7.85 (t, 2H), 7.55 (m, 1H), 7.4 (m, 2H), 3.8-3. 2 (bm, 4H), 2.7-1. 9 (bm, 4H).

The reaction mixture was stirred at 25 C for 2 hours, concentrated, and purified using reverse phase HPLC (Phenomenex Luna C8(2) 5 um IotaOmicronthetaAlpha AXIA column (30mm x 75mm) run with a gradient of acetonitrile (A) and 0.1% trifluoroacetic acid in water (B), at a flow rate of 50mL/min (0-0.5 min 10% A, 0.5-7.0 min linear gradient 10-95% A, 7.0- 10.0 min 95% A, 10.0-12.0 min linear gradient 95- 10% A) to afford the title compound.

The reaction was allowed to stir at this temperature for 2 h. The mixture was then poured into H2O and the desired product was extracted into ethyl acetate. The organic layer was backwashed once with H2O, dried over K2CO3, and concentrated under reduced vacuum to yield 416 mg as a solid.
