Preparation 4 N-(6-Amino-5-chloropyrazin-2-yl)-5-methylisoxazole-4-carboxamide Oxalyl chloride (0.8 ml, 10.3 mmol) was added to a solution of 5-Methyl-isoxazole-4-carboxylic acid (1 g, 6.9 mmol) in dichloromethane (30 ml) followed by 1 drop of dimethylformamide. The mixture was stirred at room temperature for 5 hours before concentrating in vacuo and azeotroping with dichloromethane. The residue was taken up in pyridine (3 ml) and added to a solution of 3-chloro-pyrazine-2,6-diamine (Preparation 1) (0.65 g, 4.6 mmol) in anhydrous pyridine (30 ml) and the mixture heated at 50 C. for 3 hours before cooling to room temperature and concentrating in vacuo. The residue was purified by silica gel column chromatography, eluding with ethyl acetate:heptane 1:1, to afford the product as a white solid (360 mg). 1H-NMR (d6-DMSO): 2.51 (s, 3H), 6.71 (br, s, 2H), 8.35 (s, 1H), 9.12 (s, 1H), 10.63 (br, s, 1H). MS m/z 254 [MH]+

Step A 5-Methyl-isoxazole-4-carboxylic acid [2-(4-methoxy-phenylamino)-phenyl]-amide To N-(4-methoxy-phenyl)-benzene-1,2-diamine di-hydrochloride salt (0.942 g, 3.28 mmol), 5-methyl-isoxazole-4-carboxylic acid (0.500 g, 3.93 mmol), Et3N (4.6 ml, 32.8 mmol) and DMAP. (cat.) in CH2Cl2 (10 ml) was added PPAA as a 50% solution in EtOAc (3.0 ml, 4.92 mmol). The reaction was stirred at room temperature overnight, diluted with EtOAc (100 ml) and washed with sat. NaHCO3 (2*30 ml) and brine (1*30 ml), dried (MgSO4), filtered and concentrated by vacuum. The resultant oil was subjected to flash chromatography (SiO2, biotage, 25% EtOAc/hexanes) to give 5-methyl-isoxazole-4-carboxylic acid [2-(4-methoxy-phenylamino)-phenyl]-amide as an oil (0.729 g, 2.26 mmol, 69%). MS 324 (M+1).

N-(10H-Pyrazino[2,3-b][1,4]benzothiazin-8-ylmethyl)-5-methyl-1,2-oxazole-4-carboxamide A solution of 520 mg of 5-methyl-1,2-oxazole-4-carboxylic acid and 0.75 ml of triethylamine in tetrahydrofuran (10 ml) was ice-cooled. After adding 0.8 ml of diethyl chlorophosphate, the resulting mixture was stirred at room temperature for 30 minutes. To the reaction mixture was added 5 ml of a solution of 750 mg of 8-aminomethyl-10-methoxymethyl-10H-pyrazino[2,3-b][1,4]-benzothiazine in tetrahydrofuran and the resulting mixture was stirred at room temperature for 1.5 hours. Next, the reaction mixture was distributed into ethyl acetate and an aqueous solution of ammonium chloride. The organic layer was extracted, washed with water and dried over anhydrous sodium sulfate. After distilling off the solvent under reduced pressure, the residue was purified by silica gel column chromatography (eluted with dichloromethane/methanol) to thereby give 520 mg of N-(10-methoxymethyl-10H-pyrazino[2,3-b][1,4]benzothiazin-8-ylmethyl)-5-methyl-1,2-oxazole-4-carboxamide. Further, the product was treated by the same method as the one of Example 434 to thereby give 310 mg of the title compound as yellow crystals. 1H-NMR(DMSO-d6) delta ppm: 2.63(s, 3H), 4.23(d, J=6.3 Hz, 2H), 6.71(s, 1H), 6.72(d, J=7.5 Hz, 1H), 6.86(d, J=7.5 Hz, 1H), 7.62(s, 2H), 8.79(t, J=6.3 Hz, 1H), 8.89(s, 1H), 9.50(s, 1H)
