Synthesis and Antibacterial Activity of 3-(Morpholinopyridyl)-5-substituted Isoxazole Derivatives

A series of 3-(morpholinopyridyl)-5-substituted isoxazole derivatives were prepared starting from 6-chloropyridine-3-carbaldehyde. The structures of the products were confirmed by IR, NMR and MS data. The products have an isoxazole mother core with 3-morpholinopyridyl substitution and possess an approximately planar structure. In addition a number of ester groups, amino groups, triazole groups and oxazolidinone groups were introduced to 5-position of the isoxazole mother core. The antibacterial activities of the products against Staphylococcus aureus, methicillin-resistant Staphylococcus aureus, Staphylococcus epidermis, Enterococcus faecalis and Escherichia coli were studied. Inferior activities (MIC>32 mg/L) were observed in comparison with linezolid. The results imply that the lack of 5-sp(3) hybridization structure may result in the decrease of antibacterial activity of isoxazole derivatives.

SYNTHESIS AND THERMOTROPIC PROPERTIES OF NEW MESOGENIC PYRAZOLE AND ISOXAZOLE DERIVATIVES

The synthesis of new mesogenic alkyloxy pyrazole and isoxazole derivatives is reported. The mesogenic properties of these compounds were investigated by differential scanning calorimetry, polarizing microscopy and X-ray diffraction. Almost all compounds show preferred orientation in a magnetic field of about 1.5 T. Pyrazole derivatives show higher transition temperatures than the corresponding isoxazole derivatives. Small differences (1-3 angstrom) between the molecular lengths (as determined from models and the smectic layer thickness indicated by X-rays) are explained in terms of interdigitation or skewing of the alkyloxy chains.

A library of isoxazole and 1,2,4-oxadiazole-containing diheterocyclic compounds has been prepared. Our strategy was explored in solution phase first as follows. Synthesis, Structures, and Herbicidal Activity of Isoxazole Derivatives

The several novel 3-substituted phenyl isoxazole derivatives were prepared from substituted phenyl-butan-1,3-dione. Their structures were confirmed by element analysis, IR, MS, and H-1 NMR. X-ray structure analysis indicated that the dihedral angles of the phenyl ring with the isoxazole ring in compounds 4a and 4b were 19.46 degrees and 49.18 degrees, respectively. Preliminary bioassay showed that the title compounds had good activity to various weeds. and 4a exhibited almost the same activity with 4b. This was different from the former works, which showed that the big dihedral angle of the phenyl ring with the heterocyclic moiety was necessary for high-herbicidal activity.

Condensation of beta-diketone derivative with hydroxylamine hydrochloride (NH2OH.HCl) in pyridine results in the synthesis of isoxazole derivative.
