An Expeditious and Facile Synthesis of Isoxazole Derivatives in Deep Eutectic Solvent

A convenient synthesis of (3-(3-phenyl-1-(substituted phenyl)-1H-pyrazol-4-yl)isoxazol-5-yl) methanol from pyrazole aldehyde oxime using choline chloride:glycerol (1:2) as a deep eutectic solvent has been accomplished successfully. This protocol afforded corresponding isoxazole in good to moderate yield, avoiding the use of toxic catalyst and volatile solvents. The synthesized compounds were structurally characterized on the basis of infrared, H-1-nuclear magnetic resonance (NMR), C-13-NMR, and mass analyses. The final compounds were tested for antimicrobial activities using the agar well diffusion method and the compounds containing methyl, methoxy, and nitro substituents were found to be more active.

Probes/inhibitors 2b, 3a, 3c, and 3f exerted the antiproliferative and neuroprotective activities at micromolar concentrations through inhibition of nuclear HDACs, indicating that they are cell permeable and the presence of an azide or a diazide group does not interfere with the neuroprotection properties, or enhance cellular cytotoxicity, or affect cell permeability.

Reactive organogels based on isoxazole esters: alkali metal ions selective gelation and crystallization

A series of simple ester molecules containing an isoxazole moiety were found to form instant organogels at room temperature in the presence of NaOH without the heating-cooling cycle used for conventional supramolecular gels. The gelation process was triggered due to the hydrolysis of the isoxazole esters and occurred selectively with Na+. When LiOH, NaOH and KOH were separately introduced into the methanol solutions of the isoxazole esters, the solution remained as a solution, transformed to a organogel and a crystal, respectively. With the help of a study on the phase behavior of the corresponding isoxazole acid in the presence of the alkali bases, it was revealed that pi-pi stacking of the isoxazole moiety and the ionic interaction between the carboxylates and Na+ are the main driving forces for the self-assembly and the organogelation. The size of the alkali metal ions will subtly affect the gelation, with the Li+ and K+ ions leading to solution and crystallization, respectively. These results have provided an insight into the balance between the solution, gelation and crystallization with subtle molecular variations.

Synthesis and fungicidal activities of 3-(alpha-alkoxyiminobenzyl)isoxazole derivatives

A series of 3-(alpha-alkoxyiminobenzyl)isoxazole derivatives were synthesized and their fungicidal activities against crop diseases were assessed. Studies of the structure-activity relationships revealed the strongest fungicidal activity when the alkoxyimino moiety was substituted with a methyl group. When the position-2 of the benzene ring on the benzyl moiety was substituted with a phenoxymethyl group, a good fungicidal activity was obtained. Among the compounds examined, 3-[2-(2,5-dimethylphenoxymethyl)-alpha-methoxyiminobenzyl] isoxazole (27) showed potent fungicidal activity against cucumber powdery mildew, wheat powdery mildew and wheat eye spot.
