Synthesis of novel isoxazole ring containing quinazoline derivatives and in vitro inhibitory activity against PTP alpha and PTP epsilon

7-Nitro-4-chrolo-quinazoline (4) was synthesized by two-step reactions using 4-nitro-2-aminobenezic acid and formamidine acetate as starting materials. And then, 3-(substituted-phenyl)isoxazole-5-methylamine derivatives 5a similar to 5e were synthesized by synthesizing substituted benaldehyde oxime, [3+2] dipolar cycloaddition reaction with propargyl alcohol, then acylation with methanesulfonyl chloride, substitution with NaN(3) and reduction with Zn/NH(4)Cl. Subsequently, 3-(substituted-phenyl)isoxazole-5-methylamino-4(3H)-quinazoline derivatives 6a similar to 6e were obtained by solid-grinding 5a similar to 5e and 4 under the basic Al(2)O(3) at room temperature. Their structures were confirmed by IR, HNMR and ESI-MS spectra. The compounds 6a, 6c and 6d were assayed in vitro activity against PTP alpha, PTP epsilon, and showed that the tested compounds had no notable inhibitory activity in the concentration of 20 mu g/mL.

These findings have been employed to develop efficient controlled-potential (bulk) electrosyntheses of the following compounds: benzisoxazole, methylbenzo[c]isoxazole, [1,3]dioxolo[4 ‘,5 ‘,4,5]benzo[1,2-c]isoxazole, naphtho[2,3-c]isoxazole, 6-chlorobenzo[c]isoxazole, 6-methoxybenzo[c]isoxazole, 3-methyl-benzo[c]isoxazole, 3-isopropylbenzo[c]isoxazole, and 3-phenylbenzo[c]isoxazole. In addition, we have examined the use of a variety of proton donors to optimize the production of the desired product, and we have been able to recover the proton donor at the conclusion of the electrosynthesis. In each case, the synthesized product was separated by means of normal phase chromatography and identified with the aid of NMR spectros-copy, gas chromatography (GC), and gas chromatography-mass spectrometry (GC-MS). Isolated yields of the desired products range from 63 to 92 %. Moreover, our electrosyntheses are catalyst-free, environmentally green, and rapid (similar to 30 min).

UV-Laser Photochemistry of Isoxazole Isolated in a Low-Temperature Matrix

The photochemistry of matrix-isolated isoxazole, induced by narrowband tunable UV-light, was investigated by infrared spectroscopy, with the aid of MP2/6-311+ +G(d,p) calculations. The isoxazole photoreaction starts to occur upon irradiation at lambda = 240 nm, with the dominant pathway involving decomposition to ketene and hydrogen cyanide. However, upon irradiation at lambda = 221 nm, in addition to this decomposition, isoxazole was also found to isomerize into several products: 2-formyl-2H-azirine, 3-formylketenimine, 3-hydroxypropenenitrile, imidloylketene, and 3-oxopropanenitrile. The structural and spectroscopic assignment of the different photoisomerization products was achieved by additional irradiation of the lambda = 221 nm photolyzed matrix, using UV-light with lambda >= 240 nm: (i) irradiation in the 330 <= lambda <= 340 nm range induced direct transformation of 2-formyl-2H-azirine into 3-formylketenimine; (ii) irradiation with 310 <= lambda <= 318 nm light induced the hitherto unobserved transformation of 3-formylketenimine into 3-hydroxypropenenitrile and imidoylketene; (iii) irradiation with lambda = 280 nm light permits direct identification of 3-oxopropanenitrile; (iv) under lambda = 240 nm irradiation, tautomerization of 3-hydroxypropenenitrile to 3-oxopropanenitrile is observed. On the basis of these findings, a detailed mechanistic proposal for isoxazole photochemistry is presented. Efficient Synthesis and Properties of a Photochromic Diarylethene Bearing an Isoxazole Moiety

A novel photochromic diarylethene based on isoxazole moiety was synthesized and its photochromic and fluorescent properties were also investigated. The compounds exhibited remarkable photochromism, changing from colorless to red after irradiation with UV light both in solution and in PMMA film. The fluorescence had a remarkable initial increase with subsequent dramatic decrease with increasing concentration. The results indicated that the isoxazole moiety played a very important role during the process of photochromic reaction for the diarylethene derivative.
