Discovery of 4-Amino-8-quinoline Carboxamides as Novel, Submicromolar Inhibitors of NAD-Hydrolyzing Enzyme CD38

Starting from the micromolar 8-quinoline carboxamide high-throughput screening hit 1a, a systematic exploration of the structure-activity relationships (SAR) of the 4-, 6-, and 8-substituents of the quinoline ring resulted in the identification of approximately 10-100-fold more potent human CD38 inhibitors. Several of these molecules also exhibited pharmacokinetic parameters suitable for in vivo animal studies, including low clearances and decent oral bioavailability. Two of these CD38 inhibitors, 1ah and 1ai, were shown to elevate NAD tissue levels in liver and muscle in a diet-induced obese (DIO) C57BL/6 mouse model. These inhibitor tool compounds will enable further biological studies of the CD38 enzyme as well as the investigation of the therapeutic implications of NAD enhancement in disease models of abnormally low NAD.

3-Nitrene-2-formylthiophene and 3-Nitrene-2-formylfuran: Matrix Isolation, Conformation, and Rearrangement Reactions

Two new heteroarylnitrenes, 3-nitrene-2-formylthiophene (15/15?) and 3-nitrene-2-formylfuran (16/16?), in the triplet ground state have been generated in solid Ar (10.0 K) and N2 (15.0 K) matrices by the 266 nm laser photolysis of 3-azido-2-formylthiophene (13) and 3-azido-2-formylfuran (14), respectively. According to the characterization with matrix-isolation IR spectroscopy and quantum chemical calculations at the B3LYP/6-311++G(3df,3pd) level, both nitrenes exhibit two conformations depending on the orientation of the formyl groups. Upon subsequent green-light irradiation (532 nm), both the nitrenes 15/15? and 16/16? undergo ring closure to form 3,2-thienoisoxazole (17) and 3,2-furoisoxazole (18), respectively. Traces of 3-imino-4,5-dihydrothiophene-2-ketene (19), formally formed through the intramolecular 1,4-H shift in the corresponding nitrenes 15/15?, have been also identified among the laser photolysis products of the azide 13. In sharp contrast to the photochemistry, the high-vacuum flash pyrolysis (HVFP) of the azide 13 at ca. 1000 K mainly yields imino ketene in two conformations 19/19? together with traces of isoxazole 17. In addition to the reversible conformational interconversion in the imino ketene 19 ? 19?, the photoisomerization from isoxazole 17 to imino ketene 19 has also been observed. The HVFP of the azide 14 at ca. 1000 K results in complete dissociation to HCN, C2H2, CO, CO2, H2O, and N2. Unlike the recently disclosed hydrogen-atom tunneling (HAT) in the transformation from the structurally related 2-formyl phenylnitrene (2) to imino ketene 3 in a cryogenic Ar-matrix, the absence of HAT in nitrenes 15 and 16 can be reasonably explained by the higher barrier heights and also larger barrier widths in the isomerization reactions.

Mineralization of antibiotic sulfamethoxazole by photoelectro-Fenton treatment using activated carbon fiber cathode and under UVA irradiation

The mineralization of antibiotic sulfamethoxazole (SMX) of concentrations up to 300mgL-1 was examined by photoelectro-Fenton (PEF) using an activated carbon fiber (ACF) cathode with UVA (365nm) irradiation. Comparative mineralization has been studied by different methods: RuO2/Ti anodic oxidation (AO), AO in the presence of electrogenerated H2O2 (AO-H2O2), AO-H2O2 in the presence of UVA (AO-H2O2-UVA), and both the electro-Fenton (EF) and PEF processes. PEF treatment at a low applied current of 0.36A yields a faster and more complete depollution with 80% of the TOC removed after 6h of electrolysis. The higher oxidative ability of the PEF process can be attributed to the additional hydroxyl radicals (·OH) produced by the photo-Fenton reaction. The 63% mineralization in the case of EF treatment was due to the formation of short intermediates, such as carboxylic acids, which were difficult to oxidise with ·OH. In the AO-H2O2-UVA process, about 36% of the TOC was removed after 6h electrolysis, while 28% of the TOC was removed in the AO-H2O2 process. SMX is only slightly mineralized by the AO process, with only 25% of the TOC removed. HPLC-MS analysis allowed for up to six aromatic reaction products to be identified during the SMX degradation in the PEF process, mainly formed from the hydroxylation of the aromatic ring or/and isoxazole ring, accompanied by the substitution of the amine group (on aromatic cycle) or methyl group (on isoxazole ring) by ·OH. The carboxylic acids generated, including oxalic, maleic, oxamic, formic and acetic acids, were detected by ion-exclusion chromatography. The initial organic nitrogen was mainly converted into NH4+ along with a very small proportion of NO3- ion. Considering all the oxidation intermediates and end products for SMX degradation in the PEF process, a general mineralization mechanism by ·OH and UVA was proposed.

Bark depolymerization during submerged fermentation using monofloral honey, a natural mediator substitute, and integration between laccases vs. bark biopolymers, characterized by Py-GC-MS

Due to increasing waste production and disposal problems arising from synthetic polymer production, there is a critical need to substitute materials with biodegradable and renewable resources. Attempts to use laccases as a catalyst to enhance the catalytic properties of enzymes have shown them to be a promising solution for bark depolymerization. In this study, eight different fungal strains were tested for laccase enzyme production during submerged fermentation (SF), and the Pleurotus species were shown to be the best producers among the competing fungal strains. P. pulmonarius mainly produces laccase enzyme in production medium (PM) at initial conditions of pH 5.5 and 30 C. Bark depolymerization was conducted in SF and we identified polyphenols/polyaromatic compounds after four weeks when the PM was induced with 50 mg per 100 mL of each bark during the lag-phase. During SF where honey was used as a natural mediator substitute (NMS) in the PM, laccase activities were about 1.5 times higher than those found in comparable cultures without honey in the PM. These samples were analyzed by GC-MS/MS. The laccase enzyme was purified using UNO sphere Q-1 anion exchange chromatography and the molecular weight was determined to be ?50 kDa on 10% SDS-PAGE. The laccase kinetic parameters Vmax, Km, and turnover number (Kcat) were found to be 76.9 muM min-1, 909 muM and 739 min-1, respectively, from a Lineweaver-Burk plot. Furthermore, laccases are suitable for biotechnological applications that transform bark biomass into high value bark biopolymers/biochemicals. The differences observed among the identified aromatic compound MS/MS profiles were due to the utilization of two different bark species. Py-GC-MS analysis of bark showed differing effects of fungal activity on bark composition. Polyphenolics were separated in reverse-phase mode using HPLC with a pinnacle DB Biphenyl, C18 column, and UV detector. Two recognition wavelengths of 290 and 340 nm were selected to improve the separation of each single compound in monofloral honey and bark-fermented samples. This study is novel because it replaces natural mediators (NM) with monofloral honey in PM and bark materials impregnated with honey, and studies the effects of fungi-derived laccases on bark biopolymers.
