To a solution of 1.5 g of 5-methyl-3-phenyl-4-isoxazolecarboxylic acid in 50 ml of anhydrous tetrahydrofuran stirred at -78C under N2 atmosphere, 5.9 ml of a 2.5M solution of n-butyl lithium in n-hexane was added and the solution was stirred at -78C for 2 hours. Afterwards, 0.89 ml of benzyl bromide was added at -78C and the solution stirred at room temperature for 2 hours. After overnight resting, the solution was poured into water (300 ml), acidified with 1N hydrochloric acid and extracted with ethyl acetate (2 ? 200 ml). The combined organic layers were washed with water, dried (sodium sulphate) and the solvents were evaporated to dryness. The solid residue was washed with petroleum ether to give 1.82 g (84%) of the title compound as an amorphous solid.1H-NMR (200MHz, CDCl3, delta): 9.20-11.80, br, 1H, COOH; 7.10-7.90, m, 10H, phenyl CHs; 3.45, t, 2H, CH2CH2Ph; 3.10, t, 2H, CH2CH2Ph., 1136-45-4

To this solution was added a 2M NaOH solution (6.5 mL, 13.1 mmol). After 5 min. TLC showed that the reaction was complete. To the reaction mixture was added 0.5M HCl to adjust the pH to 3-4, before extracting with ethyl acetate (2¡Á75 mL). The organic extracts were combined, washed with brine, dried over sodium sulfate, and concentrated to afford 5-phenyl-isoxazole-3-carboxylic acid (57) was obtained as a white solid (1.54 g, 94%). 57: 1H NMR (500 MHz, CDCl3): delta 9.4 (broad, 1H), 7.83 (d, 2H), 7.51 (m, 3H), 6.99 (s, 1H)

To a stirred solution of (3-(4-bromophenyl)isoxazol-5-yl)methanol (AA; 0.5 g, 1.96 mmol) in CH2CI2 (15 mL) under inert atmosphere were added pyridine (0.39 mL, 4.91 mmol) and p- dimethylaminopyridine (0.024 mg, 0.19 mmol) at 0 C. After the addition of 4-nitrophenyl carbonochloridate (DK; 0.39 g, 1.93 mmol) at 0 C, the reaction mixture was warmed to RT and stirred for 12 h. The reaction was monitored by TLC. After complete consumption of the starting material, the reaction mixture was diluted with a saturated ammonium chloride solution (20 mL) and the compound was extracted with CH2CI2 (3×20 mL). The combined organic extracts were washed with water (20 mL), dried over sodium sulfate, filtered and concentrated under reduced pressure to obtain the crude. The crude was triturated with pentane (2×15 mL) to afford crude DL (710 mg) as an off-white solid. 7063-99-2, General procedure: To a stirred mixture of NaBH4 (3 equiv), 1a-e or 13a-u (1 equiv) in reflux anhydrous THF (20 ml) was added MeOH (1ml). After refluxing for futher 30 min, the reaction mixture was pouring into ice water (10 ml), and extracted with ethyl acetate (3 ¡Á 15 mL), the organic fractions were combined, washed with saturated brine (2 ¡Á 15 ml) prior to drying over anhydrous Na2SO4. After filtration and concentrate using a rotary evaporator,the residual white solid in thenext step without further purification. To a solution of the obtained solid (1 equiv) indichloromethane (20 ml) was slowly added thionyl chloride (6 equiv) and a catalytic amount of DMF at room temperature. After stirring at 40 C for 4 h, the reaction was concentrated under reduced pressure. The residue was purified by silica gel column chromatography using a mixture of petroleum ether/ethyl acetate (20:1, v/v) aseluent to afford the desired product.

Step 2: [5-(4-Bromo-phenyl)-3-methyl-isoxazol-4-yl]-carbamic acid (R)-l-phenyl-ethyl ester[00483] 5-(4-Bromo-phenyl)-3-methyl-isoxazole-4-carboxylic acid (25g, 88.7mmol) in toluene (500mL) was added triethylamine (18.5mL, 133mmol), followed by diphenylphosphoryl azide (22.1mL, 101.9mmol). (R)-(+)- 1 -Phenylethyl alcohol (11.9mL, 97.5mmol) was added, and the reaction was stirred at 75C for 2 hours. The mixture was partitioned between EtOAc and H 0 and filtered through Celite. The aqueous layer was extracted with EtOAc, and the combined organic layers were dried over MgS04, filtered, and concentrated to give the title compound.

3-(4-bromophenyl)isoxazol-5-yl)methanol (5.0 g, 19.7 mmol) synthesized in Step 1 of Preparation Example 7, and 60% sodium hydride (1 g) were added to N,N-dimethylformamide (50 mL) and the mixture was stirred for 15 min. After methyl iodide was added thereto and completion of the reaction was confirmed by TLC, extraction was carried out with water (20 mL) and ethyl acetate (100 mL). The organic layer was washed with water (50 mL¡Á2) and brine (20 mL). The organic layer was separated, dried over anhydrous magnesium sulfate, and filtered under reduced pressure to remove ethyl acetate. The residue was purified by silica gel column chromatography using ethyl acetate and hexane as a developing solvent, thus affording the title compound 3-(4-bromophenyl)-5-(methoxymethyl)isoxazole (11f). Yield: 95%.1H NMR (CDCl3, 400 MHz): 7.67 (d, 2H, J=7.2 Hz), 7.58 (d, 2H, J=7.6 Hz), 6.53 (s, 1H), 4.57 (s, 2H), and 3.45 (s, 3H).
