Agonists and Antagonists of Protease-Activated Receptor 2 Discovered within a DNA-Encoded Chemical Library Using Mutational Stabilization of the Target

The discovery of ligands via affinity-mediated selection of DNA-encoded chemical libraries is driven by the quality and concentration of the protein target. G-protein-coupled receptors (GPCRs) and other membrane-bound targets can be difficult to isolate in their functional state and at high concentrations, and therefore have been challenging for affinity-mediated selection. Here, we report a successful selection campaign against protease-activated receptor 2 (PAR2). Using a thermo-stabilized mutant of PAR2, we conducted affinity selection using our >100-billion-compound DNA-encoded library. We observed a number of putative ligands enriched upon selection, and subsequent cellular profiling revealed these ligands to comprise both agonists and antagonists. The agonist series shared structural similarity with known agonists. The antagonists were shown to bind in a novel allosteric binding site on the PAR2 protein. This report serves to demonstrate that cell-free affinity selection against GPCRs can be achieved with mutant stabilized protein targets.

Discovery of Potent Inhibitors of 11beta-Hydroxysteroid Dehydrogenase Type 1 Using a Novel Growth-Based Protocol of in Silico Screening and Optimization in CONTOUR

With the continuous progress in ultralarge virtual libraries which are readily accessible, it is of great interest to explore this large chemical space for hit identification and lead optimization using reliable structure-based approaches. In this work, a novel growth-based screening protocol has been designed and implemented in the structure-based design platform CONTOUR. The protocol was used to screen the ZINC database in silico and optimize hits to discover 11beta-HSD1 inhibitors. In contrast to molecular docking, the virtual screening process makes significant improvements in computational efficiency without losing chemical equities through partitioning 1.8 million ZINC compounds into fragments, docking fragments to form key hydrogen bonds with anchor residues, reorganizing molecules into molecular fragment trees using matched fragments and common substructures, and then regrowing molecules with the help of developed intelligent growth features inside the protein binding site to find hits. The growth-base screening approach is validated by the high hit rate. A total of 50 compounds have been selected for testing; of these, 15 hits having diverse scaffolds are found to inhibit 11beta-HSD1 with IC50 values of less than 1 muM in a biochemical enzyme assay. The best hit which exhibits an enzyme IC50 of 33 nM is further developed to a novel series of bicyclic 11beta-HSD1 inhibitors with the best inhibition of enzyme IC50 of 3.1 nM. The final lead candidate exhibits IC50 values of 7.2 and 21 nM in enzyme and adipocyte assays, respectively, displayed greater than 1000-fold of selectivity over 11beta-HSD2 and two other related hydroxysteroid dehydrogenases, and can serve as good starting points for further optimization to develop clinical candidates.

Selective Janus Kinase 2 (JAK2) Pseudokinase Ligands with a Diaminotriazole Core

Janus kinases (JAKs) are non-receptor tyrosine kinases that are essential components of the JAK-STAT signaling pathway. Associated aberrant signaling is responsible for many forms of cancer and disorders of the immune system. The present focus is on the discovery of molecules that may regulate the activity of JAK2 by selective binding to the JAK2 pseudokinase domain, JH2. Specifically, the Val617Phe mutation in JH2 stimulates the activity of the adjacent kinase domain (JH1) resulting in myeloproliferative disorders. Starting from a non-selective screening hit, we have achieved the goal of discovering molecules that preferentially bind to the ATP binding site in JH2 instead of JH1. We report the design and synthesis of the compounds and binding results for the JH1, JH2, and JH2 V617F domains, as well as five crystal structures for JH2 complexes. Testing with a selective and non-selective JH2 binder on the autophosphorylation of wild-type and V617F JAK2 is also contrasted.

Radiation-induced degradation of sulfamethoxazole in the presence of various inorganic anions

The effect of common inorganic anions (chloride, bicarbonate, carbonate, nitrate, sulfate and phosphate) on the radiation-induced degradation of sulfamethoxazole (SMX) was investigated. In the absence of anions, the removal efficiency of SMX reached 99.6% at dose of 1 kGy. However, the presence of inorganic anions had obvious influence on SMX degradation, which was dependent on their initial concentrations. Nitrate ions had inhibitive effect, which increased with increase of its initial concentration. Chloride ions showed no obvious inhibition at low concentration, while they had significant inhibition at higher concentration. For bicarbonate, carbonate and phosphate ions, they presented inhibition at low concentration, while they had enhancing effect at higher concentration. The promotion of SMX removal efficiency was observed when the initial concentration of bicarbonate, carbonate and phosphate was 50, 20 and 50 mM, respectively. Quenching experiments proved that hydroxyl radicals made a major contribution to SMX degradation in the absence of anions. However, in the presence of 50 mM of carbonate or phosphate ions, carbonate and phosphate radicals played a key role in SMX degradation. In addition, carbonate and phosphate radicals preferentially attacked the amino group and resulted in the formation of different intermediate products compared to hydroxyl radicals. The results suggested that the effect of inorganic anions on the radiation-induced degradation of sulfonamides antibiotics should be considered when radiation technology is used for the treatment of industrial wastewater.

Small-Molecule Therapeutics for Ebola Virus (EBOV) Disease Treatment

The current epidemic of Ebola virus (EBOV) hemorrhagic fever in sub-Saharan West-African countries, in terms of severity, is second only to the human immunodeficiency virus (HIV). Besides the strong pathogenicity of EBOV, the unprecedented fatality rates (up to 90 %) are partially the result of a near total absence of any proper vaccines and therapeutics to combat the EBOV epidemic. This review highlights the recent progress in identification of purely chemical, small-molecule drugs of considerable potential to be used as effective therapeutics for prevention, diagnosis, and treatment of EBOV. In particular, we profile three compounds – favipiravir, BCX-4430, and brincidofovir – identified by the World Health Organization as high-priority targets. The detailed syntheses and modes of antiviral activity of these drugs are critically discussed. The major goal of this review is to encourage the chemistry community to focus research on this area of high social impact. This review highlights the discovery, synthesis, and modes of action of three antiviral compounds currently in development against Ebola virus (EBOV) infection.

Synthesis of heterocycle-linked C-glycosyl alpha-amino acids and C-glycopeptides

Non-natural C-glycosyl alpha-amino acids are building-blocks for the co-translational modification of natural glycopeptides. These are key components of the complex machinery that operates in vital processes of all living organisms, ranging from eubacteria to eukaryotes. While a wide range of C-glycosyl amino acids have been synthesized, we have designed and prepared a small collection of a new class of these amino acids whose structure features a heterocycle ring holding the carbohydrate and the amino acid fragments. Isoxazole, triazole, tetrazole, and pyridine moieties were used as suitable linkers in this project owing to their easy construction via Huisgen cycloaddition and Hantzsch multicomponent reaction. Details on the synthesis of each family of these glycolconjugates are given below, some emphasis being given to the efficiency and generality of each method employed. The potential of these glycosyl amino acids in non-natural glycopeptide synthesis is exemplified by the coupling of a pyridine-linked derivative with two natural amino acids to give a non-natural glycotripeptide.

Coupling of coumarin scaffold with other distinct motifs has led to the design of newer compounds against breast cancer. These distinct pharmacophores possess a diverse mode of action as well as selectivity. It has been reported in the literature that coumarin hybrids possess significant potency against breast cancer by binding to various biological targets which are associated with breast cancer. Due to low toxicity profile on various organ systems, coumarin hybrids have nowadays attracted the keen attention of researchers to explore their therapeutic ability against breast cancer. Reported coumarin hybrids include coupling with isoxazole, thiazole, monastrol, chalcone, triazole, sulphonamide, triphenylethylene, benzimidazole, pyran, imidazole, stilbene, oestrogen, phenylsulphonylfuroxan, etc. In the present review, a description of various coumarin hybrid molecules has been presented along with their structural-activity relationships.
