An unprecedented application of tributyltin hydride (TBTH) as a reagent for the reductive ionic 1,5-cyclization of 3-azamuconoates (3-azahexa-2,4-dienedioates) is described. A novel 1,5-exo-trig-cyclization of 5-chloro-3-azamuconoates was used as a base for the development of an effective two-step method for the preparation of 4-hydroxy-1H-pyrrole-3-carboxylic and 3-hydroxy-1H-pyrrole-2,4-dicarboxylic acids derivatives from readily available diazo esters and 2-chloro-2H-azirines or 4-chloroisoxazoles. The experimental results, as well as the DFT calculations of the pyrroles formation, are in good agreement with the ionic mechanism involving the initial hydride attack at the CN bond of the azamuconoate followed by tin-promoted 1,5-cyclization.

Toward the discovery and development of effective modulators of alpha-synuclein amyloid aggregation

A host of human diseases, including Parkinson’s disease and Dementia with Lewy bodies, are suspected to be directly linked to protein aggregation. Amyloid protein aggregates and oligomeric intermediates of alpha-synuclein are observed in synucleinopathies and considered to be mediators of cellular toxicity. Hence, alpha-synuclein has seen as one of the leading and most compelling targets and is receiving a great deal of attention from researchers. Nevertheless, there is no neuroprotective approach directed toward Parkinson’s disease or other synucleinopathies so far. In this review, we summarize the available data concerning inhibitors of alpha-synuclein aggregation and their advancing towards clinical use. The compounds are grouped according to their chemical structures, providing respective insights into their mechanism of action, pharmacology, and pharmacokinetics. Overall, shared structure?activity elements are emerging, as well as specific binding modes related to the ability of the modulators to establish hydrophobic and hydrogen bonds interactions with the protein. In the central nervous system (CNS), neural stem cells (NSCs) differentiate into neurons, astrocytes, and oligodendrocytes-these cell lineages are considered unidirectional and irreversible under normal conditions. The introduction of a defined set of transcription factors has been shown to directly convert terminally differentiated cells into pluripotent stem cells, reinforcing the notion that preserving cellular identity is an active process. Indeed, recent studies highlight that tumor suppressor genes (TSGs) such as Ink4a/Arf and p53, control the barrier to efficient reprogramming, leaving open the question whether the same TSGs function to maintain the differentiated state. During malignancy or following brain injury, mature astrocytes have been reported to re-express neuronal genes and re-gain neurogenic potential to a certain degree, yet few studies have addressed the underlying mechanisms due to a limited number of cellular models or tools to probe this process. Here, we use a synthetic small-molecule (isoxazole) to demonstrate that highly malignant EGFRvIII-expressing Ink4a/Arf-/-; Pten-/- astrocytes downregulated their astrocyte character, re-entered the cell cycle, and upregulated neuronal gene expression. As a collateral discovery, isoxazole small-molecules blocked tumor cell proliferation in vitro, a phenotype likely coupled to activation of neuronal gene expression. Similarly, histone deacetylase inhibitors induced neuronal gene expression and morphologic changes associated with the neuronal phenotype, suggesting the involvement of epigenetic-mediated gene activation. Our study assesses the contribution of specific genetic pathways underlying the de-differentiation potential of astrocytes and uncovers a novel pharmacological tool to explore astrocyte plasticity, which may bring insight to reprogramming and anti-tumor strategies.

IR, 1H NMR and DFT studies of novel bis-Betti base derivatives of 2,6-dihydroxynaphthalene: Thermodynamic control of diastereoselectivity and configurational preference

Diastereoselectivity and configurational preference for synthetic reaction of novel bis-Betti base derivatives of 2,6-dihydroxynaphthalene were studied through density functional theory (DFT). Enthalpy, entropy and Gibbs free energy of reactions as well as stereochemistry, thermodynamic stability and spectroscopic analysis of products were investigated. The calculated 1H nuclear magnetic resonance (NMR) and infrared (IR) spectra of different configurations were compared to experimental data in order to find the preferred isomer. The diastereoselectivity of reaction was estimated through the calculated equilibrium distribution of stereoisomers. According to these results, preparation of bis-Betti bases is an exothermic process accompanied by a decrease in entropy. The calculated gas phase heats of formation of title compounds are positive. Since the energy difference between stereoisomers is quite small, the change of Gibbs free energy during the reaction favors one configuration over other possibilities. These results are in good agreement with the experimental observations.

Natural products (NPs) have a long history as a source of, and inspiration for, novel agrochemicals. Many of the existing herbicides, fungicides, and insecticides have their origins in a wide range of NPs from a variety of sources. Owing to the changing needs of agriculture, shifts in pest spectrum, development of resistance, and evolving regulatory requirements, the need for new agrochemical tools remains as critical as ever. As such, NPs continue to be an important source of models and templates for the development of new agrochemicals, demonstrated by the fact that NP models exist for many of the pest control agents that were discovered by other means. Interestingly, there appear to be distinct differences in the success of different NP sources for different pesticide uses. Although a few microbial NPs have been important starting points in recent discoveries of some insecticidal agrochemicals, historically plant sources have contributed the most to the discovery of new insecticides. In contrast, fungi have been the most important NP sources for new fungicides. Like insecticides, plant-sourced NPs have made the largest contribution to herbicide discovery. Available data on 2014 global sales and numbers of compounds in each class of pesticides indicate that the overall impact of NPs to the discovery of herbicides has been relatively modest compared to the impact observed for fungicides and insecticides. However, as new sourcing and approaches to NP discovery evolve, the impact of NPs in all agrochemical arenas will continue to expand.
