Novel Hybrid Molecules of Isoxazole Chalcone Derivatives: Synthesis and Study of In Vitro Cytotoxic Activities

Background: Now-a-days, the model of hybrid drugs has acquired recognition in medicine due to their significant role in the treatment of different health problems. Methods: We have synthesized new series of isoxazole-chalcone conjugates (14a-m) by the Claisen-Schmidt condensation of suitable substituted acetophenones with isoxazole aldehydes (12a-d). In vitro cytotoxic activity of the synthesized compounds was studied against four different selected human cancer cell lines by using sulforhodamine B (SRB) method. Results: The adopted scheme resulted in good yields of new series of isoxazole-chalcone conjugates (14a-m). Potent cytotoxic activity was observed for compounds -14a, 14b, 14e, 14i, 14j and 14k against prostate DU-145 cancer cell line. Conclusion: The observed potent cytotoxic activities were due to the presence of 3,4,5-trimethoxyphenyl group.

Synthesis and Characterization of Isoxazole-Based Energetic Plasticizer Candidates EEIN and IDN

This study presents the synthesis of ethyl 5-[(nitrooxy)methyl]isoxazole-3-carboxylate (EEIN) and isoxazole-3,5-diylbis(methylene) dinitrate (IDN). These compounds, each derived from 5-(hydroxymethyl)isoxazole-3-carboxylate, were fully characterized and their explosive sensitivity properties were determined. EEIN was isolated as a solid material with a relatively low melting temperature and IDN was isolated as a liquid with excellent energetic performance properties. In addition, we report the molecular structure of ethyl 5-[(nitrooxy)methyl]isoxazole-3-carboxylate as determined by single-crystal X-ray diffractometry. Novel ring transformation of 5-(2,3,5-tri-O-benzoyl-beta-D-ribofuranosyl)-isoxazole-4-carbaldehyde with 1,2-diaminobenzenes to 3-cyano-1,5-benzodiazepine C-nucleosides

Syntheses of 3-cyano-7- and 8-substituted-4-(beta -D-ribofuranosyl)-1H-1,5-benzodiazepines were reported. Treatment of isoxazole carbaldehyde with 1,2-diamino-4-nitrobenzene in chloroform gave a Schiffs base, 4-(2-amino-5-nitrophenyl)iminomethyl-5-(2,3,5-tri-O-benzoyl-beta -D-ribofuranosyl)isoxazole in 82% yield with no trace of the other regioisomer. The cyclocondensation of the resulting Schiffs base in benzene containing trifluoroacetic acid (TFA) gave 3-cyano-8-nitro-4-(2,3,5-tri-O-benzoyl-beta -D-rbofuranosyl)-1H-1,5-benzodiazepine in 49% yield. The same reac tion of isoxazole carbaldeyde with 1,2-diamino-4-methoxy- and 4-chlorobenzenes afforded the corresponding Schiffs bases. Extending the reaction time for Schiff’s base gave the corresponding cyanobenzodiazepines in good yields. Debenzoylation of the compounds with sodium methoxide produced deprotected C-nucleosides.
