[Figure not available: see fulltext.] N,N-Dialkylamides and esters of 2-(chloro/bromo/iodo)-2H-azirine-2-carboxylic acids were synthesized by isomerization of 4-halo-5-(dialkylamino/alkoxy)isoxazoles in the presence of catalytic amounts of FeSO4¡¤7H2O. The use of iron(II) sulfate as catalyst, compared to its chloride, provides the advantage of avoiding halide exchange products in isomerization reactions of 4-bromo- and 4-iodoisoxazoles, as well as prevents catalyst deactivation by the 5-amino substituent of isoxazole, compared to rhodium(II) carboxylates.

Synthesis and Biological Activities of Novel Pyrazole Amide Derivatives Containing Substituted Isoxazole Group

In order to find novel pyrazole compounds possessing potent biological activities, a series of novel pyrazole amide derivatives bearing substituted isoxazole moiety were designed and synthesized according to the method of active substructure combination. Their structures were confirmed by 1H NMR, 13C NMR and elemental analysis. The bioassay data indicated that some title compounds had good insecticidal activities. At the concentration of 500 mug/mL, some compounds showed 60%~100% insecticidal activity against Oriental armyworm, and 50%~100% insecticidal activity against Aphis medicaginis. Additionally, some compounds exhibited good anti-cancer activity against HepG2 cells with the IC50 values of 14.2~18.9 mumol/L.

The syntheses and characterizations of two new energetic compounds based on a bis-isoxazole scaffold are presented. The syntheses employ a metal-free [3+2] cycloaddition of an alkyne and a nitrile oxide. In addition, we report the molecular structure of these compounds as determined by single crystal x-ray diffractometry. Both materials have favourable sensitivity properties and low melting points. Their energetic properties indicate potential use as propellant plasticizers.

Compound 11 did not display indications of animal toxicity and is currently slated for further preclinical development.

The generation of gold carbenes via the gold-catalyzed intermolecular reaction of nucleophiles containing relatively labile N-O or N-N bonds with alkynes has received considerable attention during recent years. However, this protocol is not atom-economic as the reaction produces a stoichiometric amount of pyridine or quinoline waste, the cleaved part of the N-O or N-N bonds. In this article, we disclose an unprecedented gold-catalyzed formal [3+2] cycloaddition between ynamides and isoxazoles, allowing rapid and practical access to a wide range of synthetically-useful 2-aminopyrroles. Most importantly, mechanistic studies and theoretical calculations revealed that this reaction presumably proceeds via an alpha-imino gold carbene pathway, thus providing a strategically novel, atom-economic route to the generation of gold carbenes. Other significant features of this approach include the use of readily-available starting materials, high flexibility, simple procedure, mild reaction conditions, and in particular, no need to exclude moisture or air (“open flask”).

5-Substituted 4-isoxazolecarboxamides with platelet antiaggregating and other activities

The synthesis of a series of 5-substituted 4-isoxazolecarboxamides by reaction of eight 5-substituted 4-isoxazolecarbonyl chlorides with pyrrolidine, piperidine, morpholine and 4-trifluoromethylaniline is described.
