Solar photocatalytic degradation of sulfamethoxazole by tio2 modified with noble metals

Application of solar photocatalysis for water treatment is intensively studied. In this work, we investigated TiO2 modified with platinum (Pt/TiO2) and palladium (Pd/TiO2) using sulfamethoxazole (SMX) as the model contaminant. We considered the following parameters: (i) level of TiO2 modification with Pt/Pd, (ii) initial concentration of photocatalysts, (iii) geographic location where processes were conducted, and (iv) natural water matrix. The catalysts characterized by SEM, EDX, DRS, and XRD techniques showed successful deposition of Pd and Pt atoms on TiO2 surface that enabled light absorption in the visible (Vis) range, and therefore caused efficient SMX removal in all tested conditions. A comparison of the rate constants of SMX degradation in various conditions revealed that modification with Pd gave better results than modification with Pt, which was explained by the better optical properties of Pd/TiO2. The removal of SMX was higher with Pd/TiO2 than with Pt/TiO2, independent of the modification level. In the experiments with the same modification level, similar rate constants were achieved when four times the lower concentration of Pd/TiO2 was used as compared with Pt/TiO2. Formation of four SMX transformation products was confirmed, in which both amine groups are involved in photocatalytic oxidation. No toxic effect of post-reaction solutions towards Lepidium sativum was observed. View Full-Text

N-H insertion reactions of rhodium carbenoids. Part 3. The development of a modified Bischler indole synthesis and a new protecting-group strategy for indoles

A modified version of the Bischler indole synthesis has been developed in which the key step is the N-H insertion reaction of rhodium carbene intermediates derived from alpha-diazo-beta-ketoesters with anilines. Thus N-methylanilines 1 react with diazoketoesters 2 in the presence of dirhodium(II) acetate to give (N-arylamino)ketones 3, cyclisation of which using boron trifluoride-ethyl acetate or acidic ion exchange resin gives the indoles 4. In order to extend this method to the synthesis of N-unsubstituted indoles, a new protecting group strategy for indoles was developed. In this, anilines are reacted with alpha,beta-unsaturated-esters or -sulfones to give the conjugate addition products 6 and 9, cyclisation of which gives indoles 8 and 11. The N-(2-ethoxycarbonylethyl)- and -(2-sulfonylethyl)- protecting groups are readily removed from indoles 8 and 11 by treatment with base.

Structural trends among second-generation voltage-gated sodium channel blockers

Publisher Summary The chapter examines the second-generation voltage-gated sodium (Nav) channel blockers and patent applications of the past decade and highlights structural trends by clustering compounds with similar structures. The term ‘second generation’ refers to compounds identified in the search for Nav subtype selectivity, as opposed to compounds whose Nav activity was characterized subsequent to their use in the clinic. Nav channels are transmembrane proteins that control the flow of sodium ions across cell membranes. They are important regulators of cellular excitability, contributing to the initiation and propagation of action potentials in neurons, heart and muscle. The term ‘structural trends’ refers to two-dimensional substructure motifs shared between compounds. This chapter discusses small-molecule Nav channel blockers. Blockers act as channel modulators, stabilizing one of the non-conducting states. An examination of structural trends among Nav channel blockers showed that the structure-activity relationships (SAR) are subtle and unpredictable. Small structural changes switch the selectivity between TTX-sensitive and TTX-resistant members, however these changes are often highly scaffold specific. Further developments in understanding the fine structure of Nav channels will help to rationalize these findings and facilitate the next round of drug design for these clinically important targets.

Performance evaluation of waste iron shavings (Fe0) for catalytic ozonation in removal of sulfamethoxazole from municipal wastewater treatment plant effluent in a batch mode pilot plant

Feasibility of iron shavings (Fe0) as a low-cost catalyst was evaluated in catalytic ozonation for degradation of sulfamethoxazole (SMX). The characterization of the catalyst studied by a variety of techniques like scanning electron microscopy with energy dispersive spectroscopy (SEM-EDS), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR). Experimental results indicated a first order kinetics for Fe-based catalytic ozonation (O3/Fe0) and sole-ozonation (O3) in the removal of SMX with slightly greater rate constant for O3. In the both processes, the removal efficiency of SMX was more than 99% at the first 5 min. Average COD removal efficiency in the O3/Fe0 process was about 1.36 times greater than O3. The main intermediates detected by LC-MS/MS were 3-Amino-5-methylisoxazole, benzoquinone, sulfanilic acid, hydroxyl-SMX and nitroso SMX. The mineralization of SMX significantly increased concentration of sulfate and nitrate. Fe0 enhanced ozone utilization and transfer rate. Fe0 is not requiring any preparation before use and could be a promising catalyst for practical advanced wastewater treatment.

Discovery of 4-Amino-8-quinoline Carboxamides as Novel, Submicromolar Inhibitors of NAD-Hydrolyzing Enzyme CD38

Starting from the micromolar 8-quinoline carboxamide high-throughput screening hit 1a, a systematic exploration of the structure-activity relationships (SAR) of the 4-, 6-, and 8-substituents of the quinoline ring resulted in the identification of approximately 10-100-fold more potent human CD38 inhibitors. Several of these molecules also exhibited pharmacokinetic parameters suitable for in vivo animal studies, including low clearances and decent oral bioavailability. Two of these CD38 inhibitors, 1ah and 1ai, were shown to elevate NAD tissue levels in liver and muscle in a diet-induced obese (DIO) C57BL/6 mouse model. These inhibitor tool compounds will enable further biological studies of the CD38 enzyme as well as the investigation of the therapeutic implications of NAD enhancement in disease models of abnormally low NAD.

Quantum Chemical Treatment of Molecules in Condensed Disordered Media

A short overview of the quantum chemical and statistical physical methods of modelling the solvent effects in condensed disordered media is presented. In particular, the methods for the calculation of the electrostatic, dispersion and cavity formation contributions to the solvation energy of electroneutral solutes are considered. The calculated solvation free energies, proceeding from different geometrical shapes for the solute cavity are compared with the experimental data. The self-consistent reaction field theory has been used for a correct prediction of the tautomeric equilibrium constant of acetylacetone in different dielectric media,. Finally, solvent effects on the molecular geometry and charge distribution in condensed media are discussed.
