Human proteasomes are validated targets in oncology and are promising targets in immune related diseases. Pyridin-2-ones have previously been used as constrained peptide isosteres capable to act as a hydrogen-bond acceptor. Our results demonstrate that 5-methylpyridin-2-one can be introduced into proteasome inhibitors, but that the activity of the resulting compounds is compromised compared to their parent peptide vinyl sulfones and peptide epoxy ketones. Proteasome inhibitors have reached the clinic for the treatment of multiple myeloma and mantle cell lymphoma. Most proteasome inhibitors are based on an oligopeptide scaffold. With the aim to establish whether peptidomimetics are feasible elements in the development of potentially physiologically more tractable inhibitors we studied the use of 5-methylpyridin-2-one as a peptide isoster in peptide vinyl sulfones and peptide epoxy ketones.

We have described a method for oxidative cross-coupling reactions between N-alkyl anilines and terminal alkynes forming N-aryl-N-methylpropargylamines. Superparamagnetic CuFe2O4 nanoparticles were employed as efficient and robust catalyst. The optimum conditions involved the use of tert-butyl hydroperoxide (TBHP) as oxidant and methylating reagent in dimethylacetamide (DMA) solvent at 140 C. The two-step reaction, methylation and C-C cross-coupling, proceeds efficiently and has high selectivity, and good conversions were achieved in short reaction times. Preliminary mechanistic investigation was conducted. The CuFe2O4 nanoparticles could be facilely separated from the reaction mixture by magnetic decantation and could be reused several times with only a slight decrease in catalytic activity.

In this study, a new series of 4-(5-methylisoxazol-3-ylamino)thiazole derivatives incorporated with different heterocyclic moieties was synthesized and screened for their in vitro antitumor activity against human HCT-116, HePG-2 and MCF-7 cell lines. The most promising compound 3was subjected to further kinase assay study against a panel of twelve protein kinases and exhibited good EGFR inhibitory activity (95%) and moderate PI3K (p110b/p85a) and p38alphainhibitory activities (89, 85%) respectively. Molecular modeling simulation was done to explore the binding mode of the ligand 3 within active sites EGFR, PI3K (p110b/p85a) and p38alpha kinases.
