Detoxification and degradation of sulfamethoxazole by soybean peroxidase and UV?+?H2O2 remediation approaches

Development of efficient methods for the degradation of emerging pollutants such as pharmaceuticals and pesticides are being recognized as a research priority as more and more of these organic pollutants are being detected in our water bodies. In this work, the degradation of the emerging pollutant sulfamethoxazole (SMX) was carried out using a soybean peroxidase (SBP) enzyme system and an advanced oxidation process (UV + H2O2). The optimized conditions for sulfamethoxazole degradation by the peroxidase enzyme showed an absolute requirement for a redox mediator (1-hydroxybenzotriazole) and low pH. The other conditions for an efficient degradation were as follows: [H2O2] = 56 muM, [SBP] = 78 nM and [SMX] = 5 ppm. The degradation of the pollutant was followed using UV?Vis spectrophotometry and liquid chromatography?mass spectroscopy (LC?MS). The formed products were identified using liquid chromatography?tandem mass spectrometry. The two remediation methods, an enzymatic approach using SBP + H2O2, and the photolytic technique using UV + H2O2, generated diverse sets of intermediates, suggesting that different degradation routes were at work in the two systems. Phytotoxicity studies carried out using Lactuca sativa (lettuce) showed different levels of detoxification of the SMX pollutant after the two different remediation treatments.

Herbicidal activity and molecular docking study of novel accase inhibitors

Acetyl-CoA carboxylase (ACCase) is an important target enzyme for the development of new bleaching herbicides. On the basis of structure-activity relationships and active subunit combinations, a series of novel 2-phenyl-3-cyclohexanedione enol ester derivatives was designed and synthesized by coupling and acylation reactions. The preliminary biological tests indicated good post-emergent herbicidal activity at a dosage of 150?300 g ai/ha, superior to that of clethodim against barnyard grass. Compound 3d was safe with respect to maize, even at a dosage of 300 g ai/ha. Compound 3d showed the best ACCase inhibitory activity in vitro, with a value of 0.061 nmol h-1 mg-1 protein, superior to that of clethodim. Molecular docking modeling showed that compound 3d and clethodim had the same interactions with surrounding residues, leading to an excellent combination with the active pocket of ACCase. That may have been the mechanism responsible for the death of the barnyard grass. The present work suggests compound 3d as a potential lead structure for further development of novel ACCase inhibitors.

Over the last three decades reactions of organic and organometallic compounds on silicon surfaces have been of great interest. This interest has been fueled by potential applications of such modification approaches to form stable coatings, to improve adhesion properties of organic and inorganic films on semiconductors, and to design suitable molecular electronics components. Despite enormous amount of work on chemistry of various compounds on silicon surfaces, the major driving force behind selective assembly and molecular ordering on reactive silicon surfaces and the preference for chemical reactivity of multifunctional compounds, have never before been a subject of a comprehensive review. As more complex molecular building blocks for multiple applications become available, there is a need to understand and quantify chemical handles on how to manipulate surface reactions in such a way that highly selective processes would take place. Classical kinetics and thermodynamics approaches to surface modification will be the main focus of this review. A large number of well-developed and well-understood reactions on silicon surfaces combined with better computational approaches to describe multiple surface reaction pathways will now allow us to predict, in many cases quantitatively, the selectivity of surface reactions in a variety of experimental conditions. In the past few years numerous examples of these approaches have been published. They provide a foundation for the general understanding and prediction of the chemical properties of a variety of multifunctional compounds. Most importantly, such predictions will be further used to optimize chemical modification processes both in a research laboratory and on the industrial scale. The current review will focus on the chemical control of the selectivity in reactions of multifunctional organic and organometallic molecules on silicon substrates. After a very brief review of the potential monofunctional candidate reactions and a summary of the experimental conditions, the balance of kinetic and thermodynamic factors will be discussed and the application and prediction of surface selectivity will be outlined. The examples of selective surface modification will be further considered on the most common silicon surfaces: Si(100) and Si(111), as well as on partially hydrogenated silicon substrates. Finally, some future directions for the development and the use of multifunctional compounds on silicon will be extended into the third dimension.

Chalcones and Five-Membered Heterocyclic Isosteres Bind to Alpha Synuclein Fibrils in Vitro

A series of chalcone and heterocyclic isosteres, in which the enone moiety was replaced with an isoxazole and pyrazole ring system, was synthesized and their affinities for alpha synuclein (Asyn), amyloid beta (Abeta), and tau fibrils were measured in vitro.
