There are also studies addressing hybrid compounds including an antioxidant moiety, which can be potentially useful in Alzheimer?s and Parkinson?s diseases and other neurodegenerative disorders. Additionally, other research works have also shown promising hybrid molecules for depression, autism and cocaine addiction. Thus, the development of molecular hybrid compounds seems to be a promising strategy in the discovery of novel therapeutic drugs.

A better understanding of the molecular mechanisms that control fibroblast plasticity and myofibroblast activation is essential to develop novel strategies to specifically target pathological cardiac fibrosis without disrupting the adaptive healing response. This review highlights the major transcriptional mediators of fibroblast origin and function in development and disease. The contribution of the fetal epicardial gene program will be discussed in the context of fibroblast origin in development and following injury, primarily focusing on Tcf21 and C/EBP. We will also highlight the major transcriptional regulatory axes that control fibroblast plasticity in the adult heart, including transforming growth factor beta (TGFbeta)/Smad signaling, the Rho/myocardin-related transcription factor (MRTF)/serum response factor (SRF) axis, and Calcineurin/transient receptor potential channel (TRP)/nuclear factor of activated T-Cell (NFAT) signaling. Finally, we will discuss recent strategies to divert the fibroblast transcriptional program in an effort to promote cardiomyocyte regeneration. This article is a part of a Special Issue entitled “Fibrosis and Myocardial Remodeling”.

The role of oxazolidine derivatives in the treatment of infectious and chronic diseases

Background: Despite being known for their antibacterial activity, the oxazolidines are molecules with wide pharmacological action and may act as anticonvulsants, anti-inflammatory and antineoplasic agents. However, such activities have been poorly explored and only two oxazolidinic derivatives hit the market until now. Therefore, this review covers the main biological activities of oxazolidines, indicating which of the classes and substituents have the best biological results as well as the synthesis methodology used to obtain them. Methods: The search for bibliographic data was made using a question focused on the oxazolidine structure and their respective activities, besides using inclusion/exclusion criteria clearly defined. The selected papers were subjected to qualitative content analysis methodology to be used in this review. Results: The oxazolidines remain excellent candidates for antibacterial, presenting three compounds in clinical testing phase (Radezolide, Cadezolide and Sutezolide), besides being a good candidate as antitubercular agents. Other less explored activities have niches with a great therapeutic potential such as the oxazolidines acting on 5-HT receptors (anticonvulsant) and Zolmitriptan (anti-migraine), and also mefloquine-oxazolidine derivatives which may act as antineoplasic and antitubercular agents. Conclusion: This review summarizes the versatility and great therapeutic potential that oxazolidines can offer, reinforcing the need for further studies and investments for this class of molecules.

Photocycloaddition of aromatic and aliphatic aldehydes to isoxazoles: Cycloaddition reactivity and stability studies

The first photocycloadditions of aromatic and aliphatic aldehydes to methylated isoxazoles are reported. The reactions lead solely to the exo-adducts with high regio- and diastereoselectivities. Ring methylation of the isoxazole substrates is crucial for high conversions and product stability. The 6-arylated bicyclic oxetanes 9a-9c were characterized by X-ray structure analyses and showed the highest thermal stabilities. All oxetanes formed from isoxazoles were highly acid-sensitive and also thermally unstable. Cleavage to the original substrates is dominant and the isoxazole derived oxetanes show type T photochromism.

Experimental gas-chromatographic data for 55 aromatic and aliphatic N-containing heterocyclic compounds and C5-C13 n-alkanes are analyzed. The polar constituents of the boiling points (Taupol) were calculated as the difference between the boiling points measured directly and the boiling points calculated from GC data obtained on a nonpolar column. Depending on the number, nature, and position of heteroatoms and alkyl groups in the rings and the structures of the molecules, the Taupol values vary from -11 to +82. The possibility of using Taupol values to estimate the physicochemical properties of compounds is discussed.

In silico investigation and biological evaluation of synthesized sulfamethoxazole derivatives

A series of 4-[4-(substitutedaryl/heteroaryldiazenyl]-N-(5-methylisoxazol-3-yl)benzene sulphonamide derivatives (4a-4f) were designed and synthesized coupling a mixture of diazotized sulfamethoxazole with six different phenolic and enolic compounds in an in situ reaction. The structural environment of synthesis of each molecule was confirmed by Fourier-transform infrared spectroscopy, proton nuclear magnetic resonance and elemental analysis. These derivatives were further screened in various biological assays in vivo for analgesic and antiinflammatory activities and in vitro for antioxidant and antimicrobial activities. When tested for analgesic activity at a dose of 50 mg/kg, compounds 4-((2-hydroxynaphthalen-1-yl)diazenyl)-N-(5-methylisoxazol-3-yl)benzene sulphonamide (4d) and 4-((4-hydroxy-5-isopropyl-2-methylphenyl)diazenyl)-N-(5-methylisoxazol-3- yl)benzene sulphonamide (4f) showed 58.33 and 57.76 % of pain inhibition, respectively. These two molecules also exhibited significant antioxidant activity at 10 and 50 mug/mul. The compound 4-[(4-hydroxy-2-oxo-2H-chromen-3-yl)diazenyl]-N-(5-methylisoxazol-3-yl)benzene sulphonamide (4a) exhibited antibacterial activity against Staphylococcus aureus resistance, Candida albicans and DescriptionCryptococcus neoformans at a concentration of 31.25 mug/ml. The analgesic action of these synthesised analogues was predicted in molecular docking experiments with a specific target protein, cyclooxgenase-2 of Mus musculus and results indicated all tested compounds to exhibit good binding interaction with the active site amino acid of the target enzyme.

Thermal Decomposition of Isoxazole. Experimental and Modeling Study

The thermal decomposition of isoxazole was studied behind reflected shocks in a pressurized-driver single-pulse shock tube over the temperature range 850-1100 K and overall densities of ca. 3*10 -5 mol/cm3.Acetonitrile and carbon monoxide are the major decomposition products, followed by hydrogen cyanide, acrylonitrile, propionitrile, and acetylene.Methane, ethylene, and ethane are produced in smaller quantities.There is no effect of large quantities of toluene (/ ca. 10) on the rate of formation of acetonitrile and carbon monoxide, indicating that the latter are formed in a unimolecular process.It is suggested that this reaction channel in the decomposition of isoxazole involves a simultaneous N-O bond cleavage in the 1-2 position, a hydrogen atom shift from position 5 to 4, and a rupture of the C(4)-C(5) bond with the removal of carbon monoxide from the ring: (1) C3H3ON -> CH3CN + CO.This process requires a very small N-O bond stretch which results in a very stiff transition structure.The rate constant for this reaction is k1 = 1011.94 exp(-44*103/RT)s-1.Since several products are formed by free-radical reactions, it is suggested that the initiation of free radicals involves the same reaction which proceeds at a much lower rate than reaction 1.
