Green synthesis, antiinflammatory and antimicrobial evaluation of novel isoxazole carboxamide derivatives

Isoxazole derivatives possess antibacterial, antiviral, anti-fungal, anti-inflammatory insecticidal activities. The novel series of isoxazole carboxamide derivatives were prepared by the various ketene dithioacetals condensed with hydroxyl amine hydrochloride, potassium hydroxide under microwave irradiation. All the synthesized compounds have been characterized by using elemental analysis, FT-IR, 1H NMR, 13C NMR spectroscopy and further supported by mass spectroscopy. Purity of all the compounds has been checked on thin layer chromatographic plate and HPLC technique. All the synthesized compounds were tested for their antibacterial and antifungal activity in vitro by broth dilution method with two Gram-positive bacteria, two Gram-negative bacteria and two fungal strains. The biological activities of the synthesized compounds have been compared with standard drugs Streptomycin and Greseofulvin. The compounds exhibited significant antibacterial and moderate antifungal activities. These compounds can be further exploited to get the potent lead compounds. The detailed synthesis antimicrobial and antiinflammatory screening of the new compounds are reported.

The bioactivation of drugs is often associated with toxicological outcomes; however, for most cases, the causal relationship between bioactivation and toxicity is not well established despite extensive research that attempts to elucidate the mechanisms leading to the formation of chemically reactive species, presumably the initial step towards adverse reactions. Due to rapid advancement in the research of cytochrome P450s (CYPs) and the prevalence of CYP involvement in the metabolic clearance of pharmaceuticals, CYP-mediated bioactivation is widely investigated and reviewed, while non-CYP-mediated bioactivation has not been emphasized. The widespread use of metabolic stability screening in drug discovery, however, has led to the identification of new chemical entities that rely on non-CYP enzymes for clearance, and the number of drugs that undergo metabolism via these enzymes has increased. Non-CYP enzymes can be divided into four general categories according to their enzymatic function, namely, oxidative, reductive, conjugative and hydrolytic. The aim of this review is to complement the existing literature on CYP-mediated metabolism by focusing on bioactivation mediated non-CYP enzymes and provide representative examples in each category.

At the same time, new trends in bromodomain inhibitor research are emerging, including an increased interest in non-BET bromodomains and a focus on drug synergy with established antitumor agents to improve chemotherapeutic efficacy. This review presents an updated view of the structure and function of bromodomains, traces the development of bromodomain inhibitors and their potential therapeutic applications, and surveys the current challenges and future directions of this vibrant new field in drug discovery.

DNA Encoded Libraries: A Visitor’s Guide

In 1992, Brenner and Lerner hypothesized that individual chemical transformations could be encoded in DNA, allowing the rapid synthesis and screening of large collections of small molecules. Since their report, huge investments into the development of the DNA encoded library (DEL) technology have enabled the acceleration of the drug discovery process especially early phase discovery undertakings such as target validation and hit identification. The origin of the diffuse interstellar bands (DIBs), one of the longest-standing mysteries of the interstellar medium (ISM), is explored within the framework of The Heterogeneous dust Evolution Model for Interstellar Solids (THEMIS). The likely nature of the DIB carriers and their evolution is here explored within the framework of the structures and sub-structures inherent to doped hydrogenated amorphous carbon grains in the ISM. Based on the natural aromatic-rich moieties (asphaltenes) recovered fromcoal and oil, the likely structure of their interstellar analogues is investigated within the context of the diffuse band problem. It is here proposed that the top-down evolution of interstellar carbonaceous grains, and, in particular, a-C(:H) nanoparticles, is at the heart of the formation and evolution of the DIB carriers and their associations with small molecules and radicals, such as C2, C3, CH and CN. It is most probable that the DIBs are carried by dehydrogenated, ionized, hetero-cyclic, olefinic and aromatic-rich moieties that form an integral part of the contiguous structure of hetero-atomdoped hydrogenated amorphous carbon nanoparticles and their daughter fragmentation products.Within this framework, it is proposed that polyene structures in all their variants could be viable DIB carrier candidates.

Influences of Hydrothermal Modification on Nitrogen Thermal Conversion of Low-Rank Coals

The influence of hydrothermal modification on thermal conversion of coal-N in low-rank coals was investigated in this paper. Hydrothermal modification has been performed using three typical Chinese low-rank coals at final modification temperatures of 200, 250, and 300 C. An analytical pyro-probe coupled to a pyrolysis-gas chromatography mass spectrometry (Py-GC/MS) system was used to study the release characteristics of nitrogen-containing compounds from raw and modified coal. In addition, a tube furnace apparatus was used to study the release characteristics of nitrogen pollutants during pyrolysis and combustion of Zhaotong raw and modified coal. Results showed that hydrothermal modification affected the content and type of nitrogen-containing compounds. Some triple bonds present in the coal structure are broken to form amides. The presence of azoles decreases sharply after modification, but it increases as the hydrothermal modification temperature increases. During tube furnace pyrolysis, the HCN yield decreases after modification; similarly, with the increase of modification temperature, NH3 is released in greater quantity after modification, but the release decreases as the modification temperature increases. NH3 may be converted from HCN through secondary reactions. Hydrothermal modification is conducive to NO emission reduction in coal combustion process; however, increasing the modification temperature restrains the NO emission to some extent.

Synthetic and medicinal prospective of structurally modified curcumins

Curcumin, a natural yellow phenolic compound, is present in various types of herbs, particularly in Turmeric, Curcuma longa Linn. (Zingiberaceae family) rhizomes. Curcumin is a polyphenolic natural compound with diverse and attractive biological activities. In the last decade curcumine and its various synthetic analogues have been prepared and evaluated for various pharmacological activities that prove it as a lead molecule against several biological targets. It is a natural antioxidant and exhibited many pharmacological activities such as anti-inflammatory, anti-microbial, anticancer, anti-Alzheimer in both preclinical and clinical studies. Moreover, Curcumin and its analogues have anti-tubercular, cardioprotective, anti-diabetic, hepatoprotective, neuroprotective, nephroprotective, antirheumatic and anti-viral activities. The substitutions of 1,6-heptadiene linkage moiety via carbonyl group sustituion and addition of heterocyclic linker; isoxazole, 1H-pyrazole, cyclopentanone, piperidin-4-one, N-methylpiperidin-4-one enhance biological activities. The structure activity relationship of various curcumin analogues is studied for medicinal purposes and it reveals that monocarbonyl linkage analogues have anticancer properties. The current review gives an insight of the history, chemistry, analogues and most interesting in vitro and in vivo studies on the biological effects of Curcumin and its analogues.
