[Figure not available: see fulltext.] Reactions of five- and six-membered cyclic beta-alkoxyvinyl alpha-keto esters and NCC-binucleophiles are described. The following binucleophiles were studied: heteroaromatic amines (pyrazoles, isoxazole, uracils, and isoquinolinone) and 2-(benzimidazolyl)acetonitrile. It was found that condensation proceeded regioselectively in the case of five-membered cyclic enone. Fused alpha-pyridine carboxylates, likely formed in situ, underwent lactonization, which resulted in six-membered lactones in moderate to excellent yields (53?91%). Only in the case of 3-unsubstituted 5-aminopyrazole, formation of gamma-pyridine carboxylate followed by cyclization to isomeric lactone was observed. On the contrary, six-membered cyclic enone was reactive only toward heterocyclic amines under optimized conditions and provided open-chain alpha-pyridine carboxylates in 40?80% yield.

Drinking water containing environmental endocrine disruptor compounds (EDCs) endangers human health, and researching the purification process of drinking water for the effective removal of EDCs is vitally important. Filtering plays a crucial role in the bio-adsorption of EDCs, but the adsorption mechanism that occurs between the EDCs and filters remains unclear. In this study, a quartz crystal microbalance (QCM) was employed to elucidate the adsorption mechanism because QCM is a label-free method that possesses high selectivity, high stability, and high sensitivity. The results indicated that a pseudo-first-order kinetic model best fits the adsorption process of four different EDCs, which included bisphenol A (BPA), estrone (E1), estradiol (E2), and sulfamethoxazole (SMZ), on silica (quartz sand), a typical filter material surface. The order of the amount of individual EDCs absorbed on the silica surface was qE2?> qE1?> qSMZ?> qBPA and related to their molecular structure, polarity, and chargeability. As the initial EDC concentration increased, the adsorbed amount of the four EDCs on the silica surface increased; however, the initial concentration had little effect on removal efficiency. The calculated Freundlich exponent (1/n) demonstrated SMZ and BPA showed a greater tendency for adsorption than E1 and E2. The mass response time on the surface of the silica gradually increased as the pH increased (from 5.5 to 8.5), indicating the adsorption rate was inhibited by the increase in pH. The addition of electrolytes shortened the mass response time of EDCs on the QCM chip. The pH and ionic strength produced no significant effects on adsorption because hydrophobicity was the primary contributor to adsorption. This study facilitated a better understanding of the interaction between EDCs and filters in water treatment.

Tetrafluoropropanol soluble, metal(II) hydrazone complexes derived from alkyl substituted isoxazole and barbituric acid were synthesized and evaluated as suitable optical recording materials for the recordable blu-ray disc system. The behaviour and relationships of the absorption, reflectivity and optical constants (complex refractive indices N = n + ik) for different metal(II) complexes in spin-coated films were discussed. In addition, the important thermal decomposition data of these metal(II) complexes from simultaneous TG-DSC analyses were presented as well. As an example of the utility of dye-based blu-ray recording media, we used the nickel(II) hydrazone complex thin film as recording layer for static optical recording performance test. Results showed that high reflectivity contrast over 45% and durable information marks (after repeatedly reading 20,000 times) could be achieved under optimum laser writing power and pulse width conditions, indicative of producing a very promising optical recording medium for blu-ray disc-recordable system.

DABCO-Mediated [4+1] Cycloaddition of beta,beta-Dihalo Peroxides with Sodium Azide toward Isoxazoles

The isoxazole skeleton is one of key structural motifs, which displays widespread applications in drugs, agriculture, and synthetic intermediates. Herein, we disclose the DABCO-mediated [4+1] cycloaddition of beta,beta-dihalo peroxides and sodium azide for synthesis of multisubstituted isoxazoles. The cycloaddition proceeds through the formation of N?C and N?O bonds. This method shows excellent regioselectivity and good scalability.

Synthesis of nitrogen and oxygen based pyrazole derivatives and its antitubercular and antimicrobial activity

Background: Pyrazole, oxazole and pyrimidine are nitrogen containing heterocycles which possess very good binding efficiency with receptor or proteins. Due to this effect it can be work as antitubercular agents with active substituents. The objective of this paper is clubbed the heterocycles to enhance their antitubercular potency. Methods: The active substituted pyrazole aldehydes were synthesized by reaction of phenyl hydrazine with different acetophenone followed by Vilsmeier-Haack formylation reaction. The isoxazole and aminopyrimidines were prepared by condensation of various chalcones with hydroxyl amine hydrochloride and guanidine hydrochloride respectively. The structures of the newly synthesized compounds were characterized by1H-NMR, Mass, IR and elemental analysis data. All the newly synthesized compounds were screened for their antibacterial activity against Gram-positive S.pyogenus and Gram negative E.coli, Paeruginosa, S.aureus and antitubercular activity against mycobacterium tuberculosis H37Rv. Results: Oxygen containing substituent in aromatic ring or heterocycles as a substituent were enhancing the antitubercular activity as compared to the other substituents. Ethoxy group present as a substituent in aromatic ring shows minimum inhibition concentration as compared to others. Conclusion: X-ray crystallographic study enhances that E-isomer formed in case of chalcones. The electron withdrawing group such as fluorine and chlorine enhancing the activity of Isoxazole and Pyridine as compare to the chalcones. Electron donating group such as ethoxy is increase the potency of the compounds at para position.

Combinatorial libraries of substrate-bound cyclic organic compounds

The invention relates to libraries of cyclic organic compounds and method of producing and assaying such libraries. According to the invention, each cyclic organic compound is constructed from a starting material in the form of a solid surface derivatized with a starting resin. Compounds are reacted with the resin to add or to form a cyclic group. The reactions are preferable carried out using a split resin procedure so that different compounds can be reacted with a plurality of subamounts so as to increase the size of the library. For example, compounds are reacted with a solid support bound starting resin to obtain a compound which includes an aldehyde functional group wherein the aldehyde compound or compounds reacted with it have substituents which are varied such that a mixture of products is obtained. The invention further relates to methods of producing combinatorial libraries of cyclic organic compounds from substrate bound compounds by cleaving the compounds from the support after synthesizing is completed and to assaying libraries of such compounds.

Rethinking drug design in the artificial intelligence era

Artificial intelligence (AI) tools are increasingly being applied in drug discovery. While some protagonists point to vast opportunities potentially offered by such tools, others remain sceptical, waiting for a clear impact to be shown in drug discovery projects. The reality is probably somewhere in-between these extremes, yet it is clear that AI is providing new challenges not only for the scientists involved but also for the biopharma industry and its established processes for discovering and developing new medicines. This article presents the views of a diverse group of international experts on the ?grand challenges? in small-molecule drug discovery with AI and the approaches to address them.
