Synthesis, spectral features and biological activity of some novel hetarylazo dyes derived from 6-amino-1,3-dimethyluracil

A series of hetarylazoaminouracil dyes were prepared by coupling of 6-amino-1,3-dimethyluracil with eight diazotized heterocyclic amines in nitrosyl sulphuric acid. The prepared azo dyes were characterized by UV-Vis, FT-IR, 13C NMR, 1H NMR spectroscopic techniques and elemental analysis. The solvatochromism of dyes was evaluated with respect to wavelength of maximum absorption (lambdamax) in seven solvents with different polarities: acetic acid, methanol, water, chloroform, acetonitrile, dimethyl sulfoxide and dimethyl formamide. The effects of acid, base and concentration of the dye on the visible absorption spectra were also reported. In addition, the antimicrobial activity of the synthesized dyes was evaluated on Escherichia coli, Bacillus subtilis, Micrococcus leuteus and Pseudomonas aeruginosa.

The bioavailability of 16a was 100% in rats and the systemic clearance and volume of distribution are higher than that of 1. In rats, intravenous 16a blocks big ET pressor responses with 30-fold greater potency than 1. After oral dosing at 3 mumol/kg, 16a displays enhanced duration relative to 1.

Impact of biogenic substrates on sulfamethoxazole biodegradation kinetics by Achromobacter denitrificans strain PR1

Pure cultures have been found to degrade pharmaceutical compounds. However, these cultures are rarely characterized kinetically at environmentally relevant concentrations. This study investigated the kinetics of sulfamethoxazole (SMX) degradation by Achromobacter denitrificans strain PR1 at a wide range of concentrations, from ng/L to mg/L, to assess the feasibility of using it for bioaugmentation purposes. Complete removal of SMX occurred for all concentrations tested, i.e., 150 mg/L, 500 mug/L, 20 mug/L, and 600 ng/L. The reaction rate coefficients (kbio) for the strain at the ng/L SMX range were: 63.4 ± 8.6, 570.1 ± 15.1 and 414.9 ± 124.2 L/gX SS·day), for tests fed without a supplemental carbon source, with acetate, and with succinate, respectively. These results were significantly higher than the value reported for non-augmented activated sludge (0.41 L/(g X SS·day) with hundreds of ng/L of SMX. The simultaneous consumption of an additional carbon source and SMX suggested that the energetic efficiency of the cells, boosted by the presence of biogenic substrates, was important in increasing the SMX degradation rate. The accumulation of 3-amino-5-methylisoxazole was observed as the only metabolite, which was found to be non-toxic. SMX inhibited the Vibrio fischeri luminescence after 5 min of contact, with EC50 values of about 53 mg/L. However, this study suggested that the strain PR1 still can degrade SMX up to 150 mg/L. The results of this work demonstrated that SMX degradation kinetics by A. denitrificans PR1 compares favorably with activated sludge and the strain is a potentially interesting organism for bioaugmentation for SMX removal from polluted waters.

N-heterocyclic derivatives of 2,4-dihydroxybenzcarbothioamide were synthesized from sulfinylbis- (2,4-dihydroxybenzenethioyl) and commercially available heterocyclic amines. The composition and chemical structures were confirmed by IR, 1H NMR, EI-MS, and elemental analysis. For the estimation of potential activity in vitro the MIC values against 15 strains of dermatophytes, yeasts, and molds were determined. The strongest fungistatic potency was found for N-5?-(3?-oxobenzfurylidyne)-2,4-dihydroxybenzcarbothioamide in relation to all tested dermatophyte strains with MIC = 0.48-0.98 mug/mL. On the basis of the spectroscopic data the influence of N-heterocyclic substitution on antimycotic activity is discussed.

Sulfamethoxazole (SMX) is one of the most widely applied sulfonamide antibiotics in the world, which is becoming a ubiquitous pollutant in the environment. In this mini-review, the microbial degradation of SMX was briefly reviewed. The performance of the conventional wastewater treatment plants in removing SMX was provided. Synthesis and antidiabetic performance of beta-amino ketone containing nabumetone moiety

We wish to report the further design and improved synthesis that resulted in two series of target molecules, TM-1 and TM-2, with remarkably simplified structures containing beta-amino ketone of discrete nabumetone moiety. These were obtained via a ‘one-pot, two-step, three-component’ protocol of Mannich reaction with yield up to 97%. A total of 28 out of 31 new compounds were characterized using 1H NMR, 13C NMR, ESI MS and HRMS techniques. Studies on their antidiabetic activities, screened in vitro at 10 mug mL-1 level, indicate that TM-2 possesses peroxisome proliferator-activated receptor activation and alpha-glucosidase inhibition activity significantly stronger than that of TM-1, and also that of the series B compounds that were previously synthesized by the group. Analysis of the structure-activity relationship points to the sulfanilamide unit as the most probable potent group of beta-amino ketone and, on the basis of which, a tangible strategy is presented for the development of new antidiabetic drugs.
