1,2,3-Triazole-Isoxazole Based Acetylcholinesterase Inhibitors: Synthesis, Biological Evaluation and Docking Study

In this work, a series of derivatives containing 1,2,3-triazole and isoxazole were synthesized. All of them were evaluated as novel dual AChE inhibitors. Most of synthesized compounds showed moderate to good inhibitory potency toward AChE. Among them, N-((1-(4-methylbenzyl)1H-1,2,3-triazol-4-yl) methyl)-5-(p-tolyl) isoxazole-3-carboxamide (5m) was the most potent AChE inhibitor, being 12-fold more potent than rivastigmine, as the reference drug. Also, molecular modeling revealed that compound 5m targeted both the catalytic anionic site (CAS) and the peripheral anionic site (PAS) of AChE.

Vacuum ultraviolet photoionization and ionic fragmentation of the isoxazole molecules

The photofragmentation of the isoxazole molecules producing ionized atomic and molecular fragments was investigated in the photon energy range of 9-32 eV, using synchrotron radiation excitation combined with ion time-of-flight spectrometry. Twenty-one well resolved cations were identified in the mass spectra of the isoxazole, and their appearance energies were determined. The yield curves of these cations were obtained in the photon energy ranges from their appearance energies up to 32 eV. Moreover, the total ion yield of isoxazole was recorded with high precision in the photon energy range of 9.9-10.5 eV. This allowed the determination of the adiabatic ionization energy of 9.96 (0.02) eV in excellent agreement with earlier spectroscopic studies. Our results show that the dissociative ionization of isoxazole starts from the ring-opening and isomerization of isoxazole, and further it follows strictly through its ionic states. Synthesis and anti-neuroinflammatory activity of N-heterocyclic analogs based on natural biphenyl-neolignan honokiol

Novel isoxazole and pyrazole analogs based on natural biphenyl-neolignan honokiol were synthesized and evaluated for their inhibitory activities against nitric oxide production in lipopolysaccharide-activated BV-2 microglial cells. The isoxazole skeleton was constructed via nitrile oxide cycloaddition from oxime 3 and pyrazole was generated by condensation of 4-chromone and alkylhydrazine. Among the analogs, 13b and 14a showed stronger inhibitory activities with IC50 values of 8.9 and 1.2 mu M, respectively, than honokiol.

DABCO-catalyzed sp(3) C-H activation: rapid access to isoxazole or coumarin-fused 3-quaternary carbon oxindoles and isoxazole-fused pyrrolidinones

A facile and efficient methodology was developed for the synthesis of isoxazole or coumarin-fused 3-quaternary carbon oxindoles and isoxazole-fused pyrrolidinones via DABCO-catalyzed sp(3) C-H activation of 5-methyl-isoxazole or 4-methylcoumarin. Furthermore, the biological activity of the isoxazole or coumarin-fused 3-quaternary carbon oxindoles 3 and 5 has been preliminarily demonstrated by in vitro evaluation against human prostate cancer cells PC-3 and human leukemia cells 1(562 by the MIT-based assays using the commercially available standard drug Cisplatin as a positive control. These results suggested that most of the compounds 3 and 5 showed considerable cytotoxicities to these two cell lines 1(562 and PC-3, and a methyl or an ethyl group of acrylates and an oxindole moiety located in the isoxazole-fused 3-quaternary carbon oxindoles 3 are beneficial for the activity. In addition, the activity of all the afforded isoxazole-fused pyrrolidinones 7 were also evaluated against Gram-positive bacteria Staphylococcus aureus (MTCC96) and Gram-negative bacteria Escherichia coli (ATCC25835) using Penicillin as a standard drug for Gram-positive organism or Streptomycin as standard drug for Gram-negative organism. The results demonstrated that most of the compounds 7 had comparable in vitro inhibitory activity against Gram-positive bacteria Staphylococcus aureus (MTCC96) with the positive control Penicillin. Through compound 2, fifteen novel S-triazolo[3,4-b]-1,3,4-thiadiazines (5a-5e), imidazolo[2,1-b]-1,3,4-thiadiazoles (9a-9e) and imidazolo[2,1-b]-1,3,4-oxadiazoles (10a-10e) containing isoxazole, which have potentially useful biological activities, were synthesized. The structures of the products were confirmed by elemental analyses and spectral analysis. And the characteristic data of IR, H-1 NMR and MS were explained reasonably.

Efficient Synthesis and Properties of a Photochromic Diarylethene Bearing Thiophene and Isoxazole Moieties

A novel photochromic diarylethene based on isoxazole moiety was synthesized and its photochromic and fluorescent properties were also investigated. This compound exhibited reversible photochromism, changing from colorless to red after irradiation with UV light both in solution and in poly-methyl methacrylate (PMMA) amorphous film. Also, it exhibited remarkable fluorescence switching in the solid state. The results showed that the diarylethene exhibited a relatively strong fluorescence switch along with the photochromism from open-ring isomer to closed-ring isomer. Using this dithienylethene 1c as optical storage was performed successfully.
