Few selective cyclooxygenase-1 (COX-1) inhibitors have been described up to now, although recent studies underlined the involvement of COX-1 in the carcinogenesis, pathogenesis of neuroinflammation, cardiovascular diseases and pain. Among the known COX-1 inhibitors none proved to be a good drug candidate, with the exception of mofezolac, that is clinically used as an analgesic drug. New selective inhibitors were very often discovered as a minor achievement during SAR investigations to discover selective COX-2 inhibitors (COXIBs). After a recognition of the new COX-1 inhibitors synthesized in the last five years, it was attempted to draw, for each chemical class, a structure which might highlight the determinant molecular features able to switch the selectivity towards the COX-1 isoform. Overall, this review could constitute a tool to a better design of novel selective COX-1 inhibitors, to be used in a disease theranostic approach targeting COX-1.

Carbon monoxide (CO) poisoning causes between 5,000-6,000 deaths per year in the US alone. The development of small molecule allosteric effectors of CO binding to hemoglobin (Hb) represents an important step toward making effective therapies for CO poisoning. To that end, we have found that the synthetic peptide IRL 2500 enhances CO release from COHb in air, but with concomitant hemolytic activity. We describe herein the design, synthesis, and biological evaluation of analogs of IRL 2500 that enhance the release of CO from COHb without hemolysis. These novel structures show improved aqueous solubility and reduced hemolytic activity and could lead the way to the development of small molecule therapeutics for the treatment of CO poisoning.

Here we describe the synthesis of quaternary alpha-aryl amino acids from enantiopure amino acid precursors by alpha-arylation without loss of stereochemical integrity. Our approach relies on the temporary formation of a second stereogenic centre in an N?-arylurea adduct12 of an imidazolidinone derivative6 of the precursor amino acid, and uses readily available enantiopure amino acids both as a precursor and as a source of asymmetry. It avoids the use of valuable transition metals, and enables arylation with electron-rich, electron-poor and heterocyclic substituents. Either enantiomer of the product can be formed from a single amino acid precursor. The method is practical and scalable, and provides the opportunity to produce alpha-arylated quaternary amino acids in multi-gram quantities.

Removal of sulfamethoxazole by electrochemically activated sulfate: Implications of chloride addition

Electrochemical oxidation is considered to be an attractive alternative to chemical oxidation for the treatment of polluted water. Given the of ability of boron-doped diamond (BDD) electrodes to generate hydroxyl radicals ([rad]OH), they are often selected for the degradation of persistent organic contaminants. Recently, BDD anodes have been demonstrated to form strong oxidants, sulfate radicals (SO4[rad]?), directly from sulfate ions. In this study, electrochemical activation of sulfate to SO4[rad]? at BDD anodes enhanced the removal of an antibiotic sulfamethoxazole (SMX). The rate of SMX oxidation was 6 times higher in sulfate anolyte compared to inert nitrate anolyte. Addition of chloride accelerated the disappearance of SMX in both anolytes due to electrochlorination. Yet, mineralization efficiency was decreased, particularly in Na2SO4 anolyte due to the scavenging of SO4[rad]? by Cl?. Electrogenerated SO4[rad]? yielded nitroso- and nitro-derivatives, which were not observed in the absence of sulfate. The peak intensities of chlorinated TPs were three orders of magnitude lower in Na2SO4 than in NaNO3 anolyte, suggesting that addition of sulfate may lower the formation of chlorinated organics. However, attention should be paid to the formation of inorganic byproducts, as the formation rates of toxic chlorate and in particular perchlorate were higher in Na2SO4 anolyte.
