The high-pressure behavior of 3,3?-biisoxazole-5,5?-bis-(methylene) dinitrate (BIDN) and 3,3?-biisoxazole-4,4?,5,5?-tetrakis-(methylene nitrate) (BITN) have been studied at room temperature to 25 GPa by Raman spectroscopy and powder X-ray diffraction. The Raman spectra, powder patterns, and calculated unit-cell volumes at select pressures show qualitative agreement with first-principles density function theory calculations. Over this pressure range, no evidence of polymorphism was observed. The lack of observed phase changes can be partially attributed to the strengthening of hydrogen bonding observed with pressure increase. These insights point to the potential importance of hydrogen bonding in potential melt-castable materials.

Probing the adsorption behavior of oxazole and isoxazole heterocyclic compounds onto B12N12 nanocluster surface in gas and aqueous mediums through DFT calculations

The adsorption of oxazole and isoxazole molecules over the pristine B12N12 nanocluster is investigated in terms of its geometric, the adsorption energies and the electronic properties using density functional theory calculations. The results indicate that the pristine cluster effectively chemisorbs with the oxazole and isoxazole molecules through their nitrogen atoms; whereas the interaction through the oxygen atoms is not considerable. The HOMO-LUMO gap of the cluster decrease by the adsorption of heterocycle rings. The role of solvent in improving the adsorption properties over the B12N12 surface is also considered. The obtained adsorption energies in aqueous medium are more negative than gas phase implied to the stability of the B12N12/ heterocycles complexes in the aqueous medium. Indeed, the interactions of some oxazole and isoxazole derivatives with this cluster are also investigated. The current study provides the knowledge about the performance of the B12N12 cluster in adsorbing oxazole and isoxazole moleculest.

Immune thrombocytopenia (ITP) is caused by production of an autoantibody to autologous platelets. ITP can be treated either by reducing platelet destruction or by increasing platelet production. Fcgamma receptor mediated phagocytosis of the opsonized blood cells is a well-accepted mechanism for the underlying pathogenesis of ITP and inhibition of this phagocytosis process with small molecules is a potential strategy for the development of drugs against ITP. A broad screen indicated that 4-methyl-1-phenyl-pyrazole derivative (1) could inhibit the phagocytosis of opsonized blood cells with weak potency. We reveal here the discovery of the polysulfide products, synthesis of various 1-phenyl-pyrazole derivatives, and the biological evaluation of pyrazole derivatives as inhibitors of phagocytosis for potential use as therapeutics for ITP. Substitution at C4 of the pyrazole moiety in the disulfide-bridged dimers influenced the potency in the increasing order of 10 ? 11 ? 16 < 19 < 20. A novel scaffold, 20 with an IC50 of 100 nM inhibiting opsonized blood cell phagocytosis was identified as a potential candidate for further studies. Confirmation of the disulfide bridge additionally provides clues for the non-thiol or non-disulfide bridge carrying ligands targeting ITP and other similar disorders. In the current work, novel derivatives of 1,2,4-triazole-5-thione rationally designed, synthesized and spectrally characterized as promising InhA inhibitors. Anti-mycobacterial potential was determined by resazurin microtiter assay using Mtb H37 Rv strain. The mechanism of action of these compounds was confirmed by InhA enzyme inhibition studies. 6b, the most active compound of the series displayed MIC of 0.19 muM in resazurin microtiter assay and InhA inhibition with IC50 of 90 nM.

Structural, spectroscopic (FT-IR, FT-Raman, NMR) and computational analysis (DOS, NBO, Fukui) of 3,5-dimethylisoxazole and 4-(chloromethyl)-3,5-dimethylisoxazole: A DFT study

The theoretical and computational analysis of two isoxazole derivatives 3,5-dimethylisoxazole and 4-chloromethyl-3,5-dimethylisoxazole were carried out along with some of the experimental evidences. The density functional theory calculations of these compounds were done with DFT/B3LYP/6-31+G(d,p) basis set using Gaussian 09 software. From the DFT calculations, the optimization geometry, vibrational analysis, electronic properties, local reactivity descriptors, natural bond orbitals, and other structural properties of the title compounds were elucidated. The chemical shifts of every C and H atom of the title compounds were calculated using Gauge Independent Atomic orbitals (GIAO) method for both proton and carbon NMR spectra. The molecular electrostatic potential of DMI has been found out and the difference in MEP on addition of chloromethyl group is also discussed. The hyperpolarizability calculations of the investigated molecules shows that the nonlinear optical activity of CDMI is greater when compared to DMI.
