Curcumin, a widely utilized flavor and coloring agent in food, has been shown to demonstrate powerful antioxidant, antitumor promoting and anti-inflammatory properties in vitro and in vivo. In the present work, synthesis of new heterocyclic derivatives based on Curcumin was studied. Compound 3 was synthesized via the reaction of furochromone carbaldehyde (1) with Curcumin (2) using pipredine as catalyst. Also, novel, 4,9-dimethoxy-5H-furo [3, 2-g] chromen-5-one derivatives 4a?d, 6a?d, 7, 8a?d, 9 and 10 were synthesized by the reactions of furochromone carbaldehyde (1) with different reagents (namely: appropriate amine 3a?d, appropriate hydrazine 5a?d, hydroxylamine hydrochloride, urea/thiourea, malononitrile, malononitrile with hydrazine hydrate). The structure of the synthesized products had been confirmed from their spectroscopic data (IR, 1H-NMR, 13C-NMR and mass spectra). In the present investigation, the newly synthesized products were screened using the MTT colorimetric assay for their in vitro inhibition capacity in two human cancer cell lines (hepatocellular carcinoma (HEPG2) and breast cancer (MCF-7) as well as the normal cell line (human normal melanocyte, HFB4) in comparison to the known anticancer drugs: 5-flurouracil and doxorubicin. The anticancer activity results indicated that the synthesized products 4c and 8b showed growth inhibition activity against HEPG2 cell line and synthesized products 4b and 8a showed growth inhibition activity against MCF-7, but with varying intensities in comparison to the known anticancer drugs, 5-flurouracil and doxorubicin. Cyclin dependent kinase 2 (CDK2), a major cell cycle protein, was identified as a potential molecular target of Curcumin. Furthermore, Curcumin induced G1 cell cycle arrest, which is regulated by CDK2 in cancer cells. Therefore, we used molecular modelling to study in silico the possible inhibitory effect of CDK2 by Curcumin derivatives as a possible mechanism of these compounds as anticancer agents. The molecular docking study revealed that compounds 4b, 8a and 8b were the most effective compounds in inhibiting CDk2, and, this result was in agreement with cytotoxicity assay.

The primary objective in the case of doping control is the detection of any trace of drug exposure, either parent drug or any of its metabolites, using the most powerful analytical methods which are generally based on chromatographic/mass spectrometric techniques. Of major concern in horseracing is the absence of a single organization regulating the anti-doping framework; instead of this, individual racing authorities provide rules and regulations often resulting in variations in the applied doping control programmes of different countries. The aim of this paper is to review the recent literature (approximately from 2012 to mid-2016) to highlight the numerous and diverse challenges faced in doping control of racing and equestrian sports, including the detection of designer drugs (anabolic steroids or stimulants) and of other emerging prohibited substances, such as peptides and noble gases in horse urine and plasma. Moreover, the application of ?omics? techniques (especially of metabolomics) deserves attention for establishing possible fingerprints of drug abuse as well as the evolution of instrumental analysis resulting a powerful ally in the fight against doping in equine sports. Permanganate is a versatile chemical oxidant, and has undergone a dramatic evolution toward deep insight into its reaction mechanism. However, the hydrogen abstraction of the N?H bond by permanganate remains unclear. We studied the permanganate oxidation of the emerging micropollutant sulfamethoxazole in acidic aqueous solution. The reaction followed autocatalytic kinetics and demonstrated first-order with respect to each reactant. The presence of HMnO4 accelerated the reaction rate, which was four orders of magnitude higher than that of MnO4?. Based on the identified products, the rate-limiting step was determined to be simple N?H bond oxidation by metal-oxo species permanganate. The mechanism was then studied computationally by density functional theory (DFT) using ammonia as the simplest model. Results showed that the N?H bond oxidation by MnO4? (32.86 kcal/mol) was a concerted mechanism similar to that of C?H bond oxidation, whereas HMnO4 oxidation of the N?H bond (10.44 kcal/mol) was a stepwise electron-proton transfer. This reminds us that coordination of Br°nsted acid could not only produce the stronger electrophile but also change the reaction mode by avoiding the bond cleavage in electron transfer process.

Two series of Schiff bases (SB), based on isoxazolines 9 a?f and isoxazoles 10 a?f, are described. 9 a?f were synthesized by a [3+2] cycloaddition 1,3-dipolar of arylnitrile oxide from p-nitrobenzaldehyde (1) and alkenes. And, subsequently oxidized to 10 a?f by MnO2. The isoxazolines and isoxazoles thus obtained were reduced to aniline derivatives and condensed with appropriated arylaldehydes. The SBs series isoxazolines 9 a?f presented a narrow mesophase range, while SBs series isoxazoles 10 a?f showed a large mesophase range. Nematic mesophase was observed for SBs with short and non-polar groups, while SmA and SmC mesophase for SBs with long and more polar terminal groups. The stability of the SBs is also dependent on the clearing temperature. For 10 a?f with high clearing temperature, decomposition induced by heat was observed during the first cycle of heating and in solutions of CDCl3. Series 9 a?f was more resistant to the thermal decomposition.

Quantitative structure-activity relationship (QSAR) and molecular docking studies have been done on 28 isoxazole and thiazole derivatives with anticonvulsant activity in subcutaneous pentylenetetrazole animal model. Physicochemical parameters obtained from PaDEL-Descriptors were utilized in the study. Parametric semi-empirical quantum technique PM3 available in Spartan 14 program was used to optimize the molecular structure of the dataset compounds. Genetic function algorithm, Modified-K-Medoid clustering, correlation analysis and multiple linear regressions were used to search for the best QSAR model. The model obtained had good statistical parameters (LOF = 0.056, R2 = 0.975, Q2 = 0.959, F3,15 = 198.058, R2(Pred.) = 0.761, PRESS = 0.058, SEE = 0.062 and cR2 P = 0.887) and can be utilized to predict the anticonvulsant activity of compounds that are within its chemical space. Molecular docking analysis showed that the studied compounds had a better binding affinity for gamma-aminobutyrate aminotransferase than vigabatrin which is a known inhibitor of gamma-aminobutyrate aminotransferase.
