Aza-boronic acids as non-beta-lactam inhibitors of AmpC-beta-lactamase

With the aim of improving the ability of non-beta-lactam inhibitors to inhibit AmpC-beta-lactamase, a series of 3-aza-phenyl-boronic acid derivatives was obtained using in parallel synthesis. The molecules were tested against Escherichia coli AmpC-beta-lactamase. The best inhibitors, 3-(2-hydroxy-naphthalen-1-ylazo)-phenyl-boronic acid (12) and 3-(2,4-dihydroxy-naphthalen-1-ylazo)-phenyl-boronic acid (14), showed apparent inhibition constant values (Ki) of 0.3 and 0.45muM and increased the potency of the semi-synthetic cephalosporin antibiotic, ceftazidime, lowering its minimum inhibitory concentration (MIC) value of 50%, against Gram-negative bacteria strains, producing high levels of AmpC-beta-lactamase.

Insights on sulfamethoxazole bio-transformation by environmental Proteobacteria isolates

Although sulfonamide residues are frequently reported as freshwaters contaminants, information on the ability of native bacteria to modify these synthetic antibiotics is scarce. Our purpose was to investigate the potential of bacteria from different aquatic environments to cleave or transform sulfamethoxazole (SMX) and infer on their ability to reduce the toxicity of this antibiotic. From a collection of about 100 Proteobacteria, 47 strains previously isolated from drinking water, surface water, and wastewater grew in the presence of 200 muMSMX, and were further studied. Out of these, 14 strains, mostly from mineral drinking water, transformed SMX into equimolar amounts of the lesser toxic derivative N4-acetyl-sulfamethoxazole. The highest percentage of SMX transformation was recorded for two strains affiliated to Pseudomonas mandelii. For P. mandelii McBPA4 higher SMX transformation rate and extent were observed in fed-batch (?8 muMSMX/h, 81%) than in batch conditions (?5 muMSMX/h, 25%), but similar specific transformation rates were found in both cultivation modes (?20 mumolSMX/gcell dry weight/h), indicating the dependence of the process on the microbial load. These results evidence that the capacity to transform synthetic antibiotics may be common among bacteria and highlight the potential of environmental bacteria in attenuating the potential adverse effects of pollution with sulfonamides.

Bifunctional Hydrogen Bonding of Imidazole with Water Explored by Rotational Spectroscopy and DFT Calculations

Laser vaporization of imidazole in the presence of an argon buffer gas has allowed the generation and isolation of two isomers of an imidazole monohydrate complex, denoted herein as imidH2O and H2Oimid, within a gas sample undergoing supersonic expansion. Imidazole and water are respectively proton-Accepting and proton-donating in imidH2O, but these roles are reversed in the H2Oimid complex. Both isomers have been characterized by chirped-pulse Fourier transform microwave spectroscopy between 7.0 and 18.5 GHz. The ground-state rotational spectra of four isotopologues of imidH2O and three isotopologues of H2Oimid have been measured. All spectra have been assigned and fitted to determine rotational (A0, B0, C0), centrifugal distortion (DJ, DJK), and nuclear quadrupole coupling constants (chiaa(N1), [chibb(N1)-chicc(N1)], chiaa(N3), and [chibb(N3)-chicc(N3)]). Structural parameters (r0 and rs) have been accurately determined from measured rotational constants for each isomer. The imidH2O complex contains a nonlinear hydrogen bond ((O-HbN3) = 172.1(26) in the experimentally determined, r0 geometry) between the pyridinic nitrogen of imidazole and a hydrogen atom of H2O. The DFT calculations find that the H2Oimid complex also contains a nonlinear hydrogen bond between the oxygen atom of water and the hydrogen attached to the pyrrolic nitrogen of imidazole ((OH1-N1) = 174.7). Two states observed in the spectrum of H2Oimid, assigned as 0- and 0+ states, confirm that large amplitude motions occur on the time scale of the molecular rotation. Density functional theory has been performed to characterize these large amplitude motions.

Approach to the library of 3-hydroxy-1,5-dihydro-2H-pyrrol-2-ones through a three-component condensation

A convenient procedure for the parallel synthesis of 3-hydroxy-1,5-dihydro- 2H-pyrrol-2-ones through a three-component condensation of active methylene compounds, aldehydes, and amines was developed.
