Synthesis, spectral and thermal properties of homo- and copolymers of 2-[(5-methylisoxazol-3-yl)amino]-2-oxo-ethyl methacrylate with styrene and methyl methacrylate and determination of monomer reactivity ratios

The methacrylate monomer, 2-[(5-methylisoxazol-3-yl)amino]-2-oxo-ethyl methacrylate (IAOEMA), was synthesized by reacting 2-chloro-N-(5-methylisoxazol)acetamide dissolved in acetonitrile with sodium methacrylate in the presence of triethylbenzylammoniumchloride (TEBAC). The free-radical-initiated copolymerization of IAOEMA, with styrene (ST) and methyl methacrylate (MMA) was carried out in dimethylsulphoxide (DMSO) solution at 65 C using 2,2?-azobisisobutyronitrile (AIBN) as an initiator with different monomer-to-monomer ratios in the feed. The monomer (IAOEMA) and copolymers were characterized by FTIR, 1H- and 13C-NMR spectral studies. The copolymer composition was evaluated by nitrogen content in polymers led to the determination of reactivity ratios. The reactivity ratios of the monomers were determined by the application of Fineman-Ross and Kelen-Tu?do?s methods. The analysis of reactivity ratios revealed that ST and MMA are more reactive than IAOEMA, and copolymers formed are statisticalle in nature. The molecular weights (Mw and Mn) and polydispersity index of the polymers were determined using gel permeation chromagtography. Glass transition temperatures of the copolymers were found to increase with an increase in the mole fraction of IAOEMA in the copolymers. The apparent thermal decomposition activation energies (E d) were calculated by Ozawa method using the SETARAM Labsys TGA thermobalance.

Janus kinases (JAKs) are non-receptor tyrosine kinases that are essential components of the JAK-STAT signaling pathway. Associated aberrant signaling is responsible for many forms of cancer and disorders of the immune system. The present focus is on the discovery of molecules that may regulate the activity of JAK2 by selective binding to the JAK2 pseudokinase domain, JH2. Specifically, the Val617Phe mutation in JH2 stimulates the activity of the adjacent kinase domain (JH1) resulting in myeloproliferative disorders. Starting from a non-selective screening hit, we have achieved the goal of discovering molecules that preferentially bind to the ATP binding site in JH2 instead of JH1. We report the design and synthesis of the compounds and binding results for the JH1, JH2, and JH2 V617F domains, as well as five crystal structures for JH2 complexes. Testing with a selective and non-selective JH2 binder on the autophosphorylation of wild-type and V617F JAK2 is also contrasted.

Radiation-induced degradation of sulfamethoxazole in the presence of various inorganic anions

The effect of common inorganic anions (chloride, bicarbonate, carbonate, nitrate, sulfate and phosphate) on the radiation-induced degradation of sulfamethoxazole (SMX) was investigated. In the absence of anions, the removal efficiency of SMX reached 99.6% at dose of 1 kGy. However, the presence of inorganic anions had obvious influence on SMX degradation, which was dependent on their initial concentrations. Nitrate ions had inhibitive effect, which increased with increase of its initial concentration. Chloride ions showed no obvious inhibition at low concentration, while they had significant inhibition at higher concentration. For bicarbonate, carbonate and phosphate ions, they presented inhibition at low concentration, while they had enhancing effect at higher concentration. The promotion of SMX removal efficiency was observed when the initial concentration of bicarbonate, carbonate and phosphate was 50, 20 and 50 mM, respectively. Quenching experiments proved that hydroxyl radicals made a major contribution to SMX degradation in the absence of anions. However, in the presence of 50 mM of carbonate or phosphate ions, carbonate and phosphate radicals played a key role in SMX degradation. In addition, carbonate and phosphate radicals preferentially attacked the amino group and resulted in the formation of different intermediate products compared to hydroxyl radicals. The results suggested that the effect of inorganic anions on the radiation-induced degradation of sulfonamides antibiotics should be considered when radiation technology is used for the treatment of industrial wastewater.

Ozonation of sulfamethoxazole promoted by MWCNT

Multi-walled carbon nanotubes (MWCNTs) with different surface chemical properties were prepared by oxidative and thermal treatments. Samples were characterized by nitrogen adsorption, temperature programmed desorption and pH at the point of zero charge. These catalysts were tested in the ozonation of sulfamethoxazole. Complete conversion of this compound was achieved after approximately 30 min. MWCNTs significantly enhanced the mineralization degree compared to single ozonation. Some oxidation products were quantified. The catalytic process is favored by MWCNTs with basic or neutral properties. Successive experimental runs of SMX degradation show that the catalysts suffer some deactivation.

New N-aryl isoxazolecarboxamides and N-isoxazolylbenzamides as anticonvulsant agents

We prepared a series of N-aryl isoxazolecarboxamide, N-isoxazolylbenzamide compounds and derivatives and studied their anticonvulsant action in MES and MMS tests. Some of these reveal considerable activity, especially with respect to MES test. The disubstitution in the 2.6-position on the phenyl ring by two methyl groups would appear to be of primary importance for the activity. The amide bridge between the phenyl and isoxazolic rings, whether of the anilide or benzamide type, seems to show similar anticonvulsant behavior. We have selected the derivatives 8 (N-(2.6-dimethylphenyl)-5-methyl-3-isoxazolecarboxamide, 12 (N-(2.6-dimethylphenyl)-5-hydroxymethyl-3-isoxazolecarboxamide) and 51 (N-(5-methyl-3-isoxazolyl)-2.6-dimethylbenzamide) which are presently being studied in more extended pharmacological tests.
