Since cyclooxygenase (COX) isozymes discovery, many papers and reviews have been published to describe the structural bases of COX inhibition, and to debate on the therapeutic and adverse effects of worldwide clinically used nonsteroidal anti-inflammatory drugs (NSAIDs), included COX-2 selective inhibitors (well known as Coxibs). COX-2 inhibition has been widely investigated, whereas the role of COX-1 in human pathophysiology is mostly not yet well ascertained. As time goes on, the cliche that the constitutively expressed isoform COX-1 is only involved in normal physiological functions, such as platelet aggregation, gastric mucosa protection and renal electrolyte homeostasis is going to be shattered. Low-dose aspirin, behaving as a preferential inhibitor of platelet COX-1, allowed to enlighten the role exerted by this isoenzyme in many mammalian cell types. This review would elucidate the most recent findings on selective COX-1 inhibition and their relevance to human pathology such as cancer, neuro-inflammation, cardioprotection, fever and pain. It would also focus on the design and development of new highly selective COX-1 inhibitors, useful tools in pharmacological studies aimed at gaining a deeper insight of the role of COX-1 in human health and disease. Among the traditional NSAIDs, other then aspirin and indomethacin, only few examples of selective COX-1 inhibitors (SC-560, FR122047, mofezolac, P6 and TFAP) have been so far identified. This review has also the scope to stimulate the development of novel drugs, which activity is COX-1 mediated.

Enantioenriched heteroaryl ethanols and aryl heteroarylmethanols are important intermediates and structural motifs in medicinal chemistry. Asymmetric biocatalytic reduction of corresponding ketones provides a straightforward approach for preparation of these compounds. Accordingly, three newly isolated fungal strains have been described, which produced the desired heteroaryl alcohols in high enantiomeric excess (ee). A broad substrate specificity was observed within these limited number of biocatalysts as demonstrated by preparation of a variety of heteroaryl alcohols, including (S)-5-(1-hydroxyethyl)furo[2,3-c]pyridine, a key intermediate for HIV-1 reverse transcriptase inhibitor, (S)-phenyl(pyridin-2-yl)methanol, an analgesic and (S,. S)-2,6-bis(1-hydroxyethyl)pyridine, a chiral building block, mostly in >99% ee and 80-92% yield. Micro-morphologically, one of the isolate was found to be similar to Penicillium funiculosum. However, its beta-tubulin sequence showed only 88% sequence identity with the known beta-tubulin sequences of Penicillium. It may, therefore, represent a new species of Penicillium. The other biocatalysts were identified as Alternaria alternata and Talaromyces flavus.

Oxidation of Pharmaceuticals by Ferrate(VI) in Hydrolyzed Urine: Effects of Major Inorganic Constituents

Destruction of pharmaceuticals excreted in urine can be an efficient approach to eliminate these environmental pollutants. However, urine contains high concentrations of chloride, ammonium, and bicarbonate, which may hinder treatment processes. This study evaluated the application of ferrate(VI) (FeVIO42-, Fe(VI)) to oxidize pharmaceuticals (carbamazepine (CBZ), naproxen (NAP), trimethoprim (TMP), and sulfonamide antibiotics (SAs)) in synthetic hydrolyzed human urine and uncovered new effects from urine’s major inorganic constituents. Chloride slightly decreased pharmaceuticals’ removal rate by Fe(VI) due to the ionic strength effect. Ammonium (0.5 M) in undiluted hydrolyzed urine posed a strong scavenging effect, but lower concentrations (?0.25 M) of ammonium enhanced the pharmaceuticals’ degradation by 300 muM Fe(VI), likely due to the reactive ammonium complex form of Fe(V)/Fe(IV). For the first time, bicarbonate was found to significantly promote the oxidation of aniline-containing SAs by Fe(VI) and alter the reaction stoichiometry of Fe(VI) and SA from 4:1 to 3:1. In depth investigation indicated that bicarbonate not only changed the Fe(VI)/SA complexation ratio from 1:2 to 1:1 but provided a stabilizing effect for Fe(V) intermediate formed in situ, enabling its degradation of SAs. Overall, the results of this study suggested that Fe(VI) is a promising oxidant for the removal of pharmaceuticals in hydrolyzed urine.

Discovery and characterization of ML398, a potent and selective antagonist of the D4 receptor with in vivo activity

Herein, we report the structure-activity relationship of a chiral morpholine-based scaffold, which led to the identification of a potent and selective dopamine 4 (D4) receptor antagonist. The 4-chlorobenzyl moiety was identified, and the compound was designated an MLPCN probe molecule, ML398. ML398 is potent against the D4 receptor with IC50 = 130 nM and Ki = 36 nM and shows no activity against the other dopamine receptors tested (>20 muM against D1, D2S, D2L, D3, and D5). Further in vivo studies showed that ML398 reversed cocaine-induced hyperlocomotion at 10 mg/kg.

Cross-coupling of a diverse range of aryl bromides with triallyl(aryl)silanes is effective in the presence of PdCl2/PCy 3 and tetrabutylammonium fluoride (TBAF) in DMSO-H2O to give various biaryls in good yields. It is worthwhile to note that the all-carbon-substituted arylsilanes, stable towards moisture, acid, and base and easily accessible, serve as a highly practical alternative to their aryl(halo)silane counterparts. A catalyst system consisting of [eta3-C3H5)PdCl]2 and 2-[2,4,6-(i-Pr)3C6Ha]-C6H4PCy 2 and use of TBAF¡¤ 3 H2O in THF-H2O are effective especially for the cross-coupling with aryl chlorides. Both of the catalyst systems tolerate a broad spectrum of common functional groups. The high efficiency of reactions is presumably due to the ready cleavage of the allyl groups upon treatment with TBAF¡¤3 H2O and an appropriate amount of water. Diallyl(diphenyl)silane also cross-couples with various aryl bromides and chlorides in good yields, whereas allyl(triphenyl)silane gives the cross-coupled products in only moderate yields. Through sequential cross-coupling of bromochlorobenzenes with different arylsilanes, a range of unsymmetrical terphenyls are accessible in good overall yields.
