The human proteome comprises 46 bromodomain-containing proteins with a total of 61 bromodomains, which, despite highly conserved structural features, recognize a wide array of natural peptide ligands. At the same time, new trends in bromodomain inhibitor research are emerging, including an increased interest in non-BET bromodomains and a focus on drug synergy with established antitumor agents to improve chemotherapeutic efficacy. This review presents an updated view of the structure and function of bromodomains, traces the development of bromodomain inhibitors and their potential therapeutic applications, and surveys the current challenges and future directions of this vibrant new field in drug discovery.

New N-aryl isoxazolecarboxamides and N-isoxazolylbenzamides as anticonvulsant agents

We prepared a series of N-aryl isoxazolecarboxamide, N-isoxazolylbenzamide compounds and derivatives and studied their anticonvulsant action in MES and MMS tests. Some of these reveal considerable activity, especially with respect to MES test. The disubstitution in the 2.6-position on the phenyl ring by two methyl groups would appear to be of primary importance for the activity. The amide bridge between the phenyl and isoxazolic rings, whether of the anilide or benzamide type, seems to show similar anticonvulsant behavior. We have selected the derivatives 8 (N-(2.6-dimethylphenyl)-5-methyl-3-isoxazolecarboxamide, 12 (N-(2.6-dimethylphenyl)-5-hydroxymethyl-3-isoxazolecarboxamide) and 51 (N-(5-methyl-3-isoxazolyl)-2.6-dimethylbenzamide) which are presently being studied in more extended pharmacological tests.

New Anti-Inflammatory Hybrid N-Acyl Hydrazone-Linked Isoxazole Derivatives as COX-2 Inhibitors:Rational Design, Synthesis and Biological Evaluation

Selectivity of a drug becomes a major problem in the treatment of a clinical disease and many nonsteroidal anti-inflammatory drugs (NSAID’s) are classic examples of this conundrum. In the present study, we designed a set of hybrid compounds (11 a-p, 12 a-i) that compose new N-acyl hydrazones (NAH) linked with isoxazoles, using rational drug design approach. The binding affinities of newly designed compounds were calculated and compared with known non anti-inflammatory NAH and cyclooxygenase-2 (COX-2) inhibitors (coxibs) at the active sites of cyclooxygenase-1 (COX-1) and COX-2. The comparison revealed that the hybrid compounds bind with COX-2 active site in a fashion quite similar to known non anti-inflammatory compounds (4). Based on these findings, we further synthesized the compounds (11 a-p, 12 a-i) and subjected them to in vitro, in vivo anti-inflammatory studies. Compound 11 l (IC50=5.8 muM) and 12 c (IC50 = 4.1 muM) were found to be active COX-2 inhibitors. Compounds 12 a, 12 b and 12 e displayed COX-2 enzyme inhibition at lower micro molar concentrations. The compounds were also evaluated for antioxidant activity and compound 11 h exhibited an effective antioxidant activity when compared with ascorbic acid. Additionally, these compounds were screened for anti-bacterial activities and compound 11 l and 12 h were exhibited greater anti-bacterial activity against gram +ve and -ve than standards Ciprofloxacin and Flucloxacillin.

Novel inhibitors of Staphylococcus aureus RnpA that synergize with mupirocin

We recently discovered RnpA as a promising new drug discovery target for methicillin-resistant S. aureus (MRSA). RnpA is an essential protein that is thought to perform two required cellular processes. As part of the RNA degrasome Rnpa mediates RNA degradation. In combination with rnpB it forms RNase P haloenzymes which are required for tRNA maturation. A high throughput screen identified RNPA2000 as an inhibitor of both RnpA-associated activities that displayed antibacterial activity against clinically relevant strains of S. aureus, including MRSA. Structure-activity studies aimed at improving potency and replacing the potentially metabotoxic furan moiety led to the identification of a number of more potent analogs. Many of these new analogs possessed overt cellular toxicity that precluded their use as antibiotics but two derivatives, including compound 5o, displayed an impressive synergy with mupirocin, an antibiotic used for decolonizing MSRA whose effectiveness has recently been jeopardized by bacterial resistance. Based on our results, compounds like 5o may ultimately find use in resensitizing mupirocin-resistant bacteria to mupirocin.
