Synthesis of functionalized isoxazole-oxindole hybrids via on water, catalyst free vinylogous Henry and 1,6-Michael addition reactions

Various 3-substituted-3-hydroxy isoxazole-oxindole hybrids were synthesized via the vinylogous Henry reaction of 3,5-dimethyl-4-nitroisoxazole and isatin using water as a reaction medium under catalyst free conditions at 50 C. Systematic studies were carried out to understand the role of the water on the reaction by using D2O, brine, ethylene glycol and PEG-400 as a reaction medium along with organic solvents. Among all of these, water (0.170 mol concentration) was found be more efficient giving desired products in 82-99% yields in 45-120 min. Further the quaternary centre (3 alcohol) generated was used for the creation of a double bond which was again used for a 1,6-Michael reaction to produce highly functionalized isoxazole-oxindole derivatives.

Design, synthesis, in vitro antimicrobial and anticancer activity of novel methylenebis-isoxazolo[4,5-b]azepines derivatives

A series of novel methylene bis-isoxazolo[4,5-b]azepines have been synthesized by reaction of 3,5-dimethyl-4-nitroisoxazole 6 with an appropriate methylene bis-chalcones 7 to obtain various Michael adducts 8a-i, which on treatment with SnCl2-MeOH underwent reductive cyclization to afford the title compounds 9a-i. Structure of these compounds were established on the basis of IR, 1H NMR, 13C NMR and mass spectral data. The title compounds 9a-i were evaluated for their in vitro antimicrobial and anticancer activities. Compounds 9h and 9i exhibited potent antimicrobial and anticancer activities as that of standard drugs.

Abstract: An efficient and solvent-free procedure for the synthesis of 3-methyl-4-nitro-5-styrylisoxazoles using nano-titania as solid support and recyclable catalyst is presented. This method provides clean, simple, solvent-free and useful alternative to synthesize styrylisoxazoles. The use of nano-titania provides excellent yield, leading to an easy separation and reuse of the catalyst up to four times without loss of yield. Also, the green matrices calculation shows low environment impact. The green chemistry matrices atom economy, reaction mass efficiency (RME), process mass intensity and E-factor are also calculated which show that this methodology is green and eco-friendly. The catalytic efficiency of heterogeneous TiO2 NPs was successfully demonstrated by recyclability experiment (up to 4 cycles). The sustainability of the catalyst was tested by performing recyclability experiment up to 4 cycles. Calculated turn over frequency (TOF) for each cycle indicates the protocol as sustainable. Graphical Abstract: SYNOPSIS: An efficient and solvent-free procedure for the synthesis of 3-methyl-4-nitro-5-styrylisoxazoles using nano-titania as solid support and recyclable catalyst is presented.
