Identification of Potential Inhibitors of the Antigen 85C of Mycobacterium tuberculosis from Natural Resources by Virtual Screening
Tuberculosis, caused by Mycobacterium tuberculosis, is one of the deadliest infections in the world and, therefore, the search for new therapeutic alternatives is strongly required. A promising etiological agent?s molecular target for drug development is one of the mycolyltransferase enzymes, the 85C antigen (Ag85C), since it is crucial for the biosynthesis of myolates, essential elements of the bacterial cell wall. In this perception, the present work aimed to identify potential inhibitors against M. tuberculosis Ag85C by hierarchical virtual screening. For this, the ZINC15 natural products database was subjected to chemical similarity screening in order to select molecules in biologically relevant space. Subsequently, molecular docking was employed to list the natural products selected in the previous step according to their affinity to the enzyme active site. The complex with the best binding affinity was submitted to Molecular Dynamics (MD) simulation with the aim of evaluating its mechanical stability. Among 100,095 evaluated compounds, the isoxazoline derivative ZINC000107283125 was top ranked (GridScore -84.65 Kcal/mol), and therefore submitted to MD simulations. According to RMSD analysis there was less fluctuation of protein residues in the HOLO form than in the APO form, which indicates that ZINC000107283125 was able to stabilize the protein structure. The observed intermolecular (protein-ligand) interactions were similar to those described in the literature, of these, we highlight the hydrogen bond with Ser125. Therefore, this study contributed for the search of new and more selective and potent drugs against Ag85C.
