Hearing impairment most often involves loss of sensory hair cells and auditory neurons. As this loss is permanent in humans, a cell therapy approach has been suggested to replace damaged cells. It is thus of interest to generate lineage restricted progenitor cells appropriate for cell based therapies. Human long-term self-renewing neuroepithelial stem (lt-NES) cell lines exhibit in vitro a developmental potency to differentiate into CNS neural lineages, and importantly lack this potency in vivo, i.e do not form teratomas. Small-molecules-driven differentiation is today an established route obtain specific cell derivatives from stem cells. In this study, we have investigated the effects of three small molecules SB431542, ISX9 and Metformin to direct differentiation of lt-NES cells into sensory neurons. Exposure of lt-NES cells to Metformin or SB431542 did not induce any marked induction of markers for sensory neurons. However, a four days exposure to the ISX9 small molecule resulted in reduced expression of NeuroD1 mRNA as well as enhanced mRNA levels of GATA3, a marker and important player in auditory neuron specification and development. Subsequent culture in the presence of the neurotrophic factors BDNF and NT3 for another seven days yielded a further increase of mRNA expression for GATA3. This regimen resulted in a frequency of up to 25-30% of cells staining positive for Brn3a/Tuj1. We conclude that an approach with ISX9 small molecule induction of lt-NES cells into auditory like neurons may thus be an attractive route for obtaining safe cell replacement therapy of sensorineural hearing loss.

Antibiotic wastewater has become a major concern due to the toxicity and recalcitrance of antibiotics. Anaerobic membrane bioreactors (AnMBRs) are considered alternative technology for treating antibiotic wastewater because of their advantages over the conventional anaerobic processes and aerobic MBRs. However, membrane fouling remains the most challenging issue in the AnMBRs? operation and this limits their application. This review critically discusses: (i) antibiotics removal and antibiotic resistance genes (ARGs) in different types of AnMBRs and the impact of antibiotics on membrane fouling and (ii) the integrated AnMBRs systems for fouling control and removal of antibiotics. The presence of antibiotics in AnMBRs could aggravate membrane fouling by influencing fouling-related factors (i.e., sludge particle size, extracellular polymeric substances (EPS), soluble microbial products (SMP), and fouling-related microbial communities). Conclusively, integrated AnMBR systems can be a practical technology for antibiotic wastewater treatment.

Improving biodegradation of pharmaceuticals during wastewater treatment is critical to control the release of emerging micropollutants to natural waters. In this study, biodegradation of six model pharmaceuticals was investigated at different initial concentrations in two discrete activated sludge systems, and moreover, the correlation was explored between the biodegradation rate and key molecular properties of the contaminants. First, the biodegradation rates of the pharmaceuticals were measured fitting a pseudo first?order kinetic model to the experimental kinetic data. The degradation rate constants (kbio) were found to negatively correlate to the initial concentration of the chemicals, indicating an inhibitory effect on the microorganisms by the pharmaceuticals. Further examinations of the rate data against the key molecular properties of the pharmaceuticals revealed, for the first time, that the electrophilicity index (omega), a measure of electrophilic power, served as a better indicator of the biodegradability and predictive parameter for the kbio than the conventional log KOW (a measure of hydrophobicity) in the two discrete aerobic activated sludge systems. However, the correlation strength (goodness?of?fit) between omega and kbio deteriorated when the reactor turned from aerobic to anoxic and anaerobic conditions, suggesting that electron transfer from pharmaceutical molecules to enzymes was inhibited when dissolved oxygen was deficit or absent. Our results show that omega can potentially serve as a straightforward and robust indicator for predicting the biodegradability of pharmaceutical in conventional activated sludge processes.

The photochemistry of 1,2-benzisoxazole (1) was studied using low-temperature matrix isolation coupled with infrared spectroscopy and quantum chemistry calculations. We identified, for the first time, spiro-2H-azirine 2 and ketenimine 3 as intermediates in the photoisomerization of 1 to 2-cyanophenol (4). These results constitute indirect evidence for the existence of vinylnitrene intermediates in the photochemistry of 1,2-benzisoxazoles. The potential energy surface (PES) resulting from the N?O bond cleavage of 1 was compared with the respective PES of the parent isoxazole. Calculations at the CBS-QB3 level show that no stabilization is gained for the triplet vinylnitrene upon introduction of a benzene ring fused with isoxazole. However, the energies of 2 and 3 are higher by 13?15 kcal/mol comparing with the 2H-azirine and ketenimine analogs resulting from isoxazole, which explains why they had not been observed before. Our general mechanistic proposal also predicts well the photoisomerizations of 2 and 3 to 4.
