A series of diversely asymmetrical mono- or disubstituted 1,2-diamidoanthraquinone derivatives were synthesized and evaluated for drug-induced cytotoxicity by SRB assay, telomerase inhibitory activity by TRAP assay, and hTERT expression by SEAP assay. Interestingly, compounds 4, 11, 21, 32 and 36 exhibited selective potent antiproliferative activities by NCI with IC50 values in the micromolar range. Of these, only compound 8 showed an IC50 value of 0.95 muM against PC-3 cell lines (human prostate cancer) by SRB assay. All the synthesized compounds exhibited a poor or modest telomerase inhibitory activity by TRAP assay suggesting another mode of action for these compounds. Compound 11 showed broad inhibition against different types of cancer cell lines in the micromolar and submicromolar range.

The kinetic study on the hydrolysis of Schiff bases (SBs) 1a-c and 2a-c induced by UV-vis was undertaken as a complementary study of the stability in solution and in bulk of the SBs. Solutions in chloroform were exposed to UV-vis and acquisition data occurred for 1a-c and for 2a-c in 21C, 30 C, 35C and 40C. Kinetic profile for 1a-c and 2a-c displayed the similar photochemical behaviour in that four temperature values. At 21C, two kinetic regimes were observed where the decomposition of SBs is faster at the initial stage, with no linear plot of absorbance vs time, and after the kinetic profile obeyed a linear behaviour. A mathematical treatment of the experimental data was applied, which allowed associating the initial stage data with a second-order reaction, and the final stage of the hydrolysis with a first-order reaction. The mechanism of photochemical hydrolysis of SBs 1a and 2a was addressed. It was composed of three parts, the excitation, then the isomerisation and activation processes of tetrahedral intermediate and, the last process, a collapse of the intermediate to the yielded products of the hydrolysis aldehydes and amines which were detected by their UV-vis spectrum.

The present study reports an asymmetric organocatalytic cascade reaction of 4-nitroisoxazole derivative with alpha,beta-unsaturated aldehydes catalysed by chiral secondary amine. Using this approach, 1,2,3-trisubstituted cyclopropane products were obtained in isolated yields up to 98% with moderate diastereoselectivities, and enantiopurity up to 99% ee. A plethora of selective COX-2 inhibitors belonging to nine chemical classes (pyrrole, imidazole, cyclopentene, benzene, pyrazole, spiroheptene, spiroheptadiene, isoxazole, and thiophene) was subjected to quantitative structure-activity relationship (QSAR) analysis using semi-empirical (AM1)-computed quantum mechanical properties and electrotopological state (E-state) indices. These computed parameters were correlated with experimental inhibitory activities (pIC50). Multilinear regression analyses produced three statistically acceptable models. Model 1 is based on quantum mechanical properties only, Model 2 is an all-E-state relationship, and Model 3 embraces both quantum mechanical and electrotopological parameters. All three models surpassed the commonly allowed minimum predictive squared correlation coefficient (q2) of 0.60. These QSAR results and the probable pharmacophore features identified in this study offer important structural insight into designing novel anti-inflammatory drugs devoid of unwelcome side effects. Guided by the generated models, 18 chemical structures belonging to spiroalkene classes were designed with calculated pIC50 values higher than that of known potent COX-2 inhibitors.

Mechanisms of magnesium homeostasis in Mycobacterium tuberculosis are poorly understood. Here, we describe the characterization of a pyrimidinetrione amide scaffold that disrupts magnesium homeostasis in the pathogen by direct binding to the CorA Mg2+/Co2+ transporter. Mutations in domains of CorA that are predicted to regulate the pore opening in response to Mg2+ ions conferred resistance to this scaffold. The pyrimidinetrione amides were cidal against the pathogen under both actively replicating and nonreplicating conditions in vitro and were efficacious against the organism during macrophage infection. However, the compound lacked efficacy in infected mice, possibly due to limited exposure. Our results indicate that inhibition of Mg2+ homeostasis by CorA is an attractive target for tuberculosis drug discovery and encourage identification of improved CorA inhibitors.
