The data in this review highlighted profens as promising lead compounds in future research to develop potent and safe anticancer agents.

Sulfamethoxazole (SMX) is an extensively consumed sulfonamide antimicrobial agent and is frequently detected in surface water. This work studied the degradation process of SMX in anodic chamber of microbial fuel cell (MFC) reactors. We found that the biodegradation of SMX could be achieved after acclimation and even high concentrations of SMX (e.g. 200 ppm) could be rapidly degraded. Excitation and emission matrix fluorescence spectroscopy analysis revealed that the chemical structure of SMX was altered during the process. Q-Exactive hybrid quadrupole-Orbitrap mass spectrometry was used to identify the degradation byproducts of SMX. The activity of electrode biofilm was examined afterwards and it was found that the microbe was in an active state. High-throughput sequencing analysis suggested that the microbial community structure was greatly changed during the process; some reported SMX scavengers, such as Achromobacter and Pseudomonas, were abundant in the reactors. Some metazoans were also recognized in the biofilm samples, which indicates that the operation of the MFC reactors was in a steady state. This study discusses the degradation mechanism of SMX and explores the microbial community response during the process, which provides useful information for the application of MFC in antibiotic elimination.

Those in rats were 920 mg/kg (p.o.), 378 mg/kg (i.p) and 572 mg/kg (s.c.) for males, and 887 mg/kg (p.o.), 342 mg/kg (i.p.) and 510 mg/kg (s.c.) for females. The sex difference was not clearly observed in mice and rats, but the species difference was observed in p.o. routes. As an initial toxic sign, the hypoactivity was observed in mice and rats of all routes, subsequently, paleness of skin, anemic conjunctiva, emaciation, stupor and/or coma were observed in mice and rats of p.o. and i.p. routes. In rats of those routes, tonic and/or asphyxial convulsion and dyspnea were also observed. A new knowledge-based scoring function (PMF-score), implemented into the DOCK4 program, was used to screen a database of 3247 small molecules for binding to the FK506 binding protein (FKBP). The computational ranking of these compounds was compared to the binding affinities measured by NMR. It was demonstrated that small, weakly binding molecules have, on average, higher computational scores than nonbinders and are enriched in the upper ranks of the computational scoring lists. In addition, the results obtained with the PMF scoring function were superior (by 30-120% larger enrichment factors) to those obtained with the standard force field score of DOCK4. The reliable ranking of small, weakly binding molecules offers new ways of designing building blocks in combinatorial libraries as well as SAR by NMR libraries with the increased chance of identifying suitable lead compounds for drug design.
