Reactivity-based dynamic covalent chemistry: Reversible binding and chirality discrimination of monoalcohols

In an effort to develop reactivity-based dynamic covalent bonding and to expand the scope and application of the dynamic covalent chemistry, in situ-generated simple generic iminium ions were utilized for the dynamic covalent binding of monoalcohols with high affinity. Hammett analysis was conducted to manipulate the equilibrium and correlate with the reactivity of reactants. The structural features of aldehydes and secondary amines were identified, and both polar and steric effects have significant impact on the binding. In particular, the substrates which can participate in pi-pi and polar-pi interactions are able to afford apparent equilibrium constants in the magnitude of 104 M-2, demonstrating the power of weak supramolecular forces to stabilize the dynamic covalent assembly. The generality of the assembly was validated with a series of mono secondary alcohols. To showcase the practicality of our system, chirality discrimination and ee measurement of chiral secondary alcohols were achieved.

Synthesis, antimicrobial, and mosquito larvicidal activity of 1-aryl-4-methyl-3,6-bis-(5-methylisoxazol-3-yl)-2-thioxo-2,3,6, 10b-tetrahydro-1H-pyrimido[5,4-c]quinolin-5-ones

A series of 1-aryl-4-methyl-3,6-bis-(5-methylisoxazol-3-yl)-2-thioxo-2,3,6, 10b-tetrahydro-1H-pyrimido[5,4-c]quinolin-5-ones (6a-h) have been synthesized by cyclization of ethyl-3-aryl-4-(2-chlorophenyl)-6-methyl-1-(5-methylisoxazol-3- yl)-2-thioxo-1,2,3,4-tetrahydropyrimidine-5-carboxylates 4a-h with 3-amino-5-methylisoxazole 5. Compounds 4a-h were obtained by Biginelli reaction, by condensation of aromatic aldehyde 1, ethyl acetoacetate 2, and isoxazolyl thioureas 3 in a one-pot reaction catalyzed by ceric ammonium nitrite (CAN). Compounds 6a-h were tested for their antibacterial and antifungal activities against various bacterial and fungal strains. The results showed that these compounds exhibited good antibacterial and antifungal activity compared with that of standard antibiotics. Mosquito larvicidal activity of the newly synthesized compounds 6a-h is also studied against fourth instar larvae Culex quinquefasciatus. Some of the compounds are proved to be lethal for mosquito larvae.

Design and synthesis of piperazine-based matrix metalloproteinase inhibitors

A new generation of cyclic matrix metalloproteinase (MMP) inhibitors derived from dl-piperazinecarboxylic acid has been described. The design involves: incorporation of hydroxamic acid as the bidentate chelating agent for catalytic Zn2+, placement of a sulfonamide group at the 1N-position of the piperazine ring to fill the S1′ pocket of the enzyme, and finally attachment of diverse functional groups at the 4N-position to optimize potency and peroral absorption. A unique combination of all three elements produced inhibitor 20 with high affinity for MMPs 1, 3, 9, and 13 (24, 18, 1.9, and 1.3 nM, respectively). X-ray crystallography data obtained for MMP-3 cocrystallized with 20 gave detailed information on key binding interactions defining an overall scaffold geometry for piperazine-based MMP inhibitors.

Continuous niobium phosphate catalysed Skraup reaction for quinoline synthesis from solketal

Solketal is derived from the reaction of acetone with glycerol, a by-product of the biodiesel industry. We report here the continuous reaction of solketal with anilines over a solid acid niobium phosphate (NbP), for the continuous generation of quinolines in the well-established Skraup reaction. This study shows that NbP can catalyse all the stages of this multistep reaction at 250 C and 10 MPa pressure, with a selectivity for quinoline of up to 60%. We found that the catalyst eventually deactivates, most probably via a combination of coking and reduction processes but nevertheless we show the promise of this approach. We demonstrate here the application of our approach to synthesize both mono- and bis-quinolines from the commodity chemical, 4,4?-methylenedianiline.

IR, 1H NMR and DFT studies of novel bis-Betti base derivatives of 2,6-dihydroxynaphthalene: Thermodynamic control of diastereoselectivity and configurational preference

Diastereoselectivity and configurational preference for synthetic reaction of novel bis-Betti base derivatives of 2,6-dihydroxynaphthalene were studied through density functional theory (DFT). Enthalpy, entropy and Gibbs free energy of reactions as well as stereochemistry, thermodynamic stability and spectroscopic analysis of products were investigated. The calculated 1H nuclear magnetic resonance (NMR) and infrared (IR) spectra of different configurations were compared to experimental data in order to find the preferred isomer. The diastereoselectivity of reaction was estimated through the calculated equilibrium distribution of stereoisomers. According to these results, preparation of bis-Betti bases is an exothermic process accompanied by a decrease in entropy. The calculated gas phase heats of formation of title compounds are positive. Since the energy difference between stereoisomers is quite small, the change of Gibbs free energy during the reaction favors one configuration over other possibilities. These results are in good agreement with the experimental observations.
